摘要
目的评价小剂量局部胰岛素联合重组人表皮生长因子(rhEGF)对烧伤大鼠创面MIP-2和MCP-1表达的影响。方法 150只成年雄性大鼠建立深II度烧伤模型后随机分为对照组,实验组A和实验组B,每组50只。对照组大鼠创面予生理盐水处理,实验组A大鼠创面予rhEGF处理,实验组B大鼠创面予小剂量局部胰岛素联合rh EGF。同时在实验的3、6、9、12、14 d 5个时点,每组大鼠随机选取10只进行实验。观察各组大鼠创面愈合率,同时通过实时定量PCR检测MIP-2和MCP-1基因的表达并采用Western blot分析MIP-2和MCP-1蛋白表达水平,了解小剂量局部胰岛素联合rh EGF对烧伤大鼠创面MIP-2和MCP-1表达的影响。结果实验组大鼠创面愈合程度和速度较对照组明显,同时实验组B较实验组A创面愈合更快,差异有统计学意义(P<0.05)。实验组大鼠MIP-2和MCP-1基因和蛋白表达量在相同时间点较对照组明显升高,且实验组A大鼠上述基因和蛋白表达量在相同时间点较实验组B明显降低,差异有统计学意义(P<0.05)。结论小剂量局部胰岛素联合rh EGF能够有效地促进烧伤大鼠创面MIP-2和MCP-1的表达,对促进创面的早期愈合意义明显。
【Objective】To investigate the influence of local application of small dose insulin combined rh EGF on MIP-2 and MCP-1 of the surface of wound in rats.【Methods】A total of 150 rats were divided into control group,experimental group A and experimental group B after deep II degree burn model was established, each group contained 50 rats. The control group was treated by normal saline. The experimental group A was treated by rh EGF while the experimental group B was treated by local application of small dose insulin combined rh EGF. 10 rats were chosen from each group for the experiment on the day 3, 6, 9, 12, 14. The healing rate of the rats was observed, and the gene and protein expressions of MIP-2 and MCP-1 were detected.【Results】The speed of wound healing in the experimental groups was faster than that in the control group, and faster in the experimental group B than in the experimental group A(P〈0.05). The genetic and protein expressions of MIP-2 and MCP-1 in the control group were lower than those in the experimental groups at the same time point. The genetic and protein expressions of MIP-2and MCP-1 in the experimental group A were lower than those in the experimental group B at the same time point.【Conclusions】Local application of small dose insulin combined rh EGF can effectively promote the expressions of MIP-2 and MCP-1 and the wound healing.
