摘要
目的探讨Bio-Oss骨粉复合骨形态发生蛋白2(BMP-2)和血管内皮生长因子(VEGF)修复骨缺损的成骨效果。方法选取SPF级SD大鼠54只,随机分为三组:单纯骨粉组、骨粉/BMP组、骨粉/BMP/VEGF组,每组18只。麻醉后在大鼠左侧胫骨前部、胫骨结节下1 cm处磨除5 mm长的骨块缺损(包括髓腔),余留后侧1/3骨皮质和腓骨起固定和支撑作用。按分组分别植入Bio-Oss骨粉、Bio-Oss骨粉复合BMP-2、Bio-Oss骨粉复合BMP-2/VEGF。术后2、4、8周每组各取6只大鼠行X线检查、HE染色、CD31免疫组化。结果 X线检查、HE染色显示术后2周时各组新骨形成极少,4、8周时骨粉/BMP/VEGF组新骨形成最多,骨粉/BMP组次之,单纯骨粉组最少。术后2、4周时,骨粉/BMP/VEGF组的CD31阳性细胞数目最多,单纯骨粉组最少,骨粉/BMP组居中,三组间比较差异具有统计学意义(P<0.01)。术后8周单纯骨粉组CD31阳性表达较前增多,但骨粉/BMP组、骨粉/BMP/VEGF组的CD31阳性表达较4周时有所下降。结论 Bio-Oss骨粉复合BMP-2和VEGF修复骨缺损的成骨效果理想,明显优于单纯应用Bio-Oss骨粉及只复合BMP-2的效果。
Objective To investigate the osteogenic effect of Bio-Oss bone composite with BMP-2 and VEGF for repairing bone de-fects. Methods 54 SD rats were randomly divided into three groups:simple Bio-Oss bone group,Bio-Oss bone/BMP group,Bio-Oss bone/BMP/VEGF groups,and each group had 18 rats. After anesthesia,bone defects about 5mm long( including the canal )were made at the front of the left tibia of rats ,under the tibial tuberosity 1 cm. The rear 1/3 of the cortical bone and fibula were retained to fix and support the left hinds. According to different groups,bone defects were implanted by Bio-Oss bone,Bio-Oss bone/BMP-2,Bio-Oss bone/BMP-2/VEGF. After 2,4,8 weeks,in each group and at each time point,six rats were underwent X-ray examination,HE staining,and CD31 immunohistochemis-try. Results The X-ray examination,and HE staining showed that the new bone formation was minimal in each group after two weeks. At 4,8 weeks,the new bone formation was more in Bio-Oss bone/BMP/VEGF group,less in Bio-Oss bone/BMP group,least in simple Bio-Oss bone group. After 2,4 weeks,the number of CD31 positive cells in Bio-Oss bone/BMP/VEGF group was more,less in Bio-Oss bone/BMP group, least in simple Bio-Oss bone group. There was a significant difference in 3 groups( P ﹤0. 01). After 8 weeks,expression of CD31 in simple Bio-Oss bone group increased comparing with at four weeks,but in Bio-Oss bone/BMP group and Bio-Oss bone/BMP/VEGF group expression of CD31 decreased. Conclusion Promoting osteogenesis and repairing bone defects are satisfactory of Bio-Oss bone/BMP-2/VEGF. The oste-ogenesis effect is much better than that of simple Bio-Oss bone and Bio-Oss bone/BMP.
