阿魏酸对小剂量辐射比格犬造血功能的影响 被引量:8

摘要 目的观察阿魏酸对γ射线全身照射比格犬造血功能的影响,考察阿魏酸对小剂量辐射比格犬的治疗作用。方法雄性比格犬30只,共分5组,每组6只,分别为照射对照组、阳性对照组、阿魏酸高、中、低剂量组,剂量分别为12、6、3mg·kg^(-1)。采用0.6 Gyγ射线全身屏蔽(屏蔽剂量率为7.95~7.944 cGy·min^(-1))连续照射3d制作小剂量辐射比格犬模型,照后即刻口服给药,以后每天给药1次,共14d,连续观察34 d;分别在照前、照后1、4、7、10、13、16、19、22、25、28、31、34d检测外周血象,并在34 d检测骨髓集落细胞、CD34^+细胞等。结果阿魏酸中剂量组照后10~19d明显升高外周白细胞、19~25 d升高淋巴细胞、10~16 d升高中性粒细胞、7~16 d升高单核细胞、10~22 d升高血小板,造模后34 d提高CD34^+细胞数,促进CFU-GM、CFU-Mix增殖。结论阿魏酸可有效改善0.6 Gyγ射线连续3d全身照射比格犬所致的造血损伤,提高骨髓造血功能,进而促进外周血液各组分恢复正常。 Objective To investigate hematopoietic function of ferulic acid(FA) on Beagle dogs irradiated with 0.6 Gy γ cobalt-60 for three consecutive days in order to evaluate the curative effect of FA on low dose radiation-injured dog models. Methods Thirty male Beagle dogs were divided into five groups:model group, positive control, and FA groups at the dose of 12, 6, 3 mg ·kg^-1 respectively. The dogs were shielding irradiated with 0.6 Gy γ for three days. Following irradiation, FA was administered intragastrically for 14 consecutive days while model group was given an equal volume of distilled water. Blood samples were collected and the number of peripheral blood ceils was counted on days 1,4, 7, 10, 13, 16, 19, 22, 25,28, 31 and 34 using a microcell counter. Clinical indexes, such as body temperature, body weight, and clinical symptoms, were observed. The ability of colonyforming unit of bone marrow cells and the percentage of CD34^+ cells in bone marrow were detected. Results FA could increase the percentage of CD34^+ cells and the ability of colony-forming unit of granulocyte macrophage. FA at the dose of 6 mg·kg^-1 could increase the white blood cells from day 10 to 19,lymphocytes from day 19 to 25, neutrophilic granulocytes from day 10 to 16 ,monocytes from day 7 to 16, and platelets from day 10 to 22. Conclusion FA could reverse hematopoietic injury induced by 0.6 Gy γ cobalt-60 of three continuous days, improve the hematopoietic function of bone marrow, and accelerate blood cell recovery
