摘要
目的通过观察复方葡萄籽胶囊对小鼠血清超氧化物歧化酶(SOD)、丙二醛(MDA),以及肝脏谷胱甘肽过氧化物酶(GSH-PX)的活性,研究复方葡萄籽胶囊对体内抗氧化作用的影响。方法无特殊病原体(SPF)级雌性昆明种小鼠50只,随机分为空白组、模型组、复方葡萄籽胶囊高、中、低剂量组,共5组,每组10只。给药组分别用195、130和65 mg/kg复方葡萄籽胶囊灌胃,空白组与模型组按体质量用一定体积双蒸水灌胃;模型组与给药组同时用10 mg/kg黄体酮注射大腿根部,每日1次,每周6次,两侧交替;背部脱毛(面积为1.5 cm×1.5 cm),置光毒仪中于320 nm波长紫外线下照射20 min,每日1次,每周6次。30 d后取肝脏,制备血清和肝匀浆,测定小鼠血清肝脏各指标。结果血清与肝脏SOD值、血清与肝脏MDA值、肝脏GSH-PX值测定显示,模型组、空白组与各给药组之间差异均有统计学意义(P<0.05);SOD与GSH-PX活力均显著提高,而MDA含量均有不同程度下降(P<0.05)。结论复方葡萄籽胶囊有良好的抗氧化作用,同时对黄体酮及紫外线诱导的小鼠黄褐斑有较好的防治作用。
Objective To investigate the effects of compound grape seed capsules on the activity of superoxide dismutase(SOD), malondialdehyde(MDA), and the liver glutathione peroxidase(GSH-PX) in the serum of mice, and study the effects of compound grape seed capsules on in vivo antioxidation. Methods Fifty female SPF level Kunming mice were randomly divided into a total of 5 groups: the control group, model group, high, medium and low-dose of compound grape seed capsules groups(n=10 each). The treatment groups were given 195, 130 and 65 mg/kg compound grape seed capsules by gavage, respectively. And the control group and the model group were given a certain volume of double distilled water according to the weight by gavage. The model group and treatment groups were in-jected with 10 mg/kg progesterone at the upper thighs once daily, 6 times a week, alternating both sides. The animals underwent back hair removal(an area of 1.5 cm×1.5 cm), and were exposed to 320 nm wavelength ultraviolet radiation for 20 min in the phototoxic instrument, once daily, 6 times a week. Then, the serum and liver homogenate were extracted and prepared from mice livers to determine the indicators in the serum and livers of mice. Results The measurements of SOD in serum and liver values, MDA in serum and liver values, GSH-PX and liver values showed that there were significant differences among the model group, control group and each treatment group( P〈0.05). The activity of SOD and GSH-PX were significantly increased in each group while the content of MDA was decreased to some extent(P〈0.05). Conclusion The compound grape seed capsules have good antioxidant effect and protective effect on progesterone and ultraviolet-induced chloasma in mice.
