木丹颗粒对痛性糖尿病周围神经病变大鼠钙通道基因表达的影响 被引量:9

Influence of mu dan granules on the calcium channel gene expression in the experimental painful diabetes peripheral neuropathy rats

摘要 目的研究木丹颗粒对痛性糖尿病周围神经病变(PDPN)大鼠钙离子通道蛋白mRNA表达的影响,探讨木丹颗粒对PDPN的治疗作用。方法雄性Wistar大鼠90只,随机取30只设为正常组,其余大鼠造模,参照Sally等方法,腹腔注射链脲佐菌素STZ(53mg/kg)制作PDPN模型,4周后,用Vonfrey纤维丝测双后足机械痛阈,痛阈明显下降,与基础值相比下降50%者为PDPN造模成功。再将在成模的54只大鼠随机分为模型组,木丹颗粒组。木丹颗粒组给予10ml/(kg·d)灌胃,分别于药物治疗后第2、4、8周末,各组随机取8只大鼠,用RT-PCR技术法检测大鼠背根神经节N型钙离子通道mRNA表达。结果与正常组比,模型组N型钙离子通道mRNA表达在各时间点呈上升趋势(P<0.05);与模型组比,木丹颗粒组N型钙离子通道mRNA表达在各时间点呈下降趋势,8周时,本组N型钙离子通道mRNA表达显著减少(P<0.05)。结论木丹颗粒可以下调PDPN大鼠N型钙离子通道蛋白mRNA表达,对PDPN有一定的治疗作用。 Objective To investigate the influence of Mudan granules on the calcium channel protein mRNA expression in the experimental pain diabetes peripheral neuropathy rats, so as to study the therapeutic effect of Mudan Granules on the pain diabetes peripheral neuropathy. Methods 90 male Wistar rats, were randomly taken 30 as the normal group, and the rest were injected STZ(53mg/kg) intraperitoneally of rats to make the painful diabetic peripheral neuropathy model. After 4 weeks, experimenter tested with double Vonfrey fiber after sufficient mechanical pain threshold, proved the pain obviously and the models were successful. 54 survived rats were randomly divided into model group, Mudan Granule group, respectively in the treated experiments for 2, 4, 8 after the weekend, each group were randomly taken 8 rats with RT - PCR technique to observe N - type calcium channel mRNA expressional changes. Results The mRNA expression of model group was in rising trends at each time point, and Mudan Granule group in each time point showed a trend of gradual decline, but in 8 w, the mRNA expression of Mudan Granule and phenytoin sodium group significantly reduced ( P 〈 0.05 ). Conclusion About the mRNA expression of N - type calcium channel in painful diabetes peripheral neuropathy, Mudan Granules showed a trend of cut, and had certain therapeutic effect on PDNP.
