摘要
以阿霉素肾病大鼠模型为研究对象,采用了血液生化学和组织病理学方法,分析了补中益气汤对阿霉素肾病的作用和可能的机制.Wistar雄性大鼠60只,随机分为对照组(n=10)、模型组(n=10)、激素组(n=10)和补中益气汤低剂量组(n=10)、中剂量组(n=10)和高剂量组(n=10).采用阿霉素二次尾静脉注射法(首次6mg/kg,二次3mg/kg)制作阿霉素肾病的大鼠模型.在阿霉素注射后7,21,42d将大鼠置于代谢笼中分别收集24h尿液,测定24h尿蛋白.末次给药后,眼眶取血检测肾功能指标,包括肌肝SCR(serum creatine)和尿素氮BUN(blood urea nitrogen);处死大鼠并观察肾组织病理改变.结果表明:补中益气汤可以显著减少尿蛋白的排泄并缓解肾皮质上皮细胞的病理学改变.这提示补中益气汤对阿霉素大鼠能够通过改善尿蛋白的排泄,缓解肾脏病理改变,从而起到对肾脏的保护作用.
The effects and potential mechanism of the Buzhong Yiqi decoction on the adriamycin ne- phropathy rat model were analyzed using blood biochemistry and histology method. In this study, 60 Spra- gue Dawley rats were divided into six groups, including the control group (nz 10), model group (nz 10), hormone group (nzl0), and Buzhong Yiqi decoction group of low close (nzl0), middle close (nz 10), and high close (nz 10). The adriamycin nephropathy rat model was made by a single intravenous tail-vein injection of adriamycin at a close of 6 mg/kg. The 24-hour urine protein(24 HUP) was measured at 7 days,21 days, and 42 clays after the injection respectively by Bradford method. After the last admistration, or- bital blood was extracted for renal function test, including the SCR fserum creatine) and BUN (blood urea nitrogen). After this, the rats were euthanized and the renal pathological changes were examined using histology method. The results showed that not only the 24 HUP decreased significantly in the Buzhong Yiqi decoction group, but also the physiological state of podocytes got obvious improvement, which in- ferred that the Buzhong Yiqi decoction played a protective role on renal disease by improving the urine pro- tein excretion.
