摘要
目的应用DNA条形码技术对鼠疫宿主动物进行分子生物学的鉴定。方法运用PCR方法检测锡林郭勒盟9种啮齿动物的149条线粒体COI基因657 bp的片段序列,用MEGA 5.0软件采用邻接法(NJ)构建分析系统发育树。结果 NJ进化树能清楚地区分9个不同的类群。结论通过COI基因应用DNA条码技术可以对锡林郭勒盟的鼠疫宿主动物进行鉴别,同时可为形态鉴别提供佐证数据。
Objective To apply the DNA barcoding in the molecular biology identification of host animals i0r plague. Methods 149 specimens of 9 species were sequenced for a 657bp region of the mitochondrial cyto- chrome oxidase subunit I gene (COI) in Xilin Gol league, the phylogenetic trees were constructed with Neigh- bor - Joining (NJ) method by using MEGAS. 0. Results A Neighbor - Joining tree showed 9 major clusters apparently. Conclusions COI sequencing of DNA barcoding can be used to identify host animals for plague in Xilin Gol league, and can provide stronger evidences for the identification of species morphology.
