自噬在黄芩素降解转染PC12细胞中α-syn过表达的作用

摘要 目的研究黄芩素是否可通过ALP通路降解转染PC12细胞内异常聚集α-syn;方法诱导自噬,采用Western Blotting法检测黄芩素对自噬-溶酶体通路中LC3-Ⅱ、p62等蛋白的表达影响。结果 20μmol·L-1黄芩素处理24h可降低p62表达,增加LC3-Ⅱ表达,诱导自噬,与对照组相比有显著性差异(P<0.01)。结论黄芩素可诱导自噬,增加异常聚集α-syn降解,可为PD治疗提供新的信号转导途径。 OBJECTIVE To determine the effect of Baicalein on overexpression of α-syn degradation in transfected PC12 cells through autophagy-lysosomal pathway( ALP); METHODS By use of fluorescene microscope to detect autophagy,then the expression of LC3-Ⅱ and p62 was examined by Western Blotting analysis. RESULTS Pretreated with 20μmol·L^- 1Baicalein for 24 hours in transfected PC12 cells,we found Baicalein decreased p62 and increased LC3-Ⅱ expression. Compared with the control group,there was significant difference( P〈 0. 01). CONCLUSION Baicalein can induce autophagy and increase the degradation of α-syn,which may provide a new signal transduction pathways for PD treatment.
