白藜芦醇对力竭运动小鼠肝损伤的保护作用 被引量:7

摘要 目的:探讨白藜芦醇对力竭运动小鼠肝损伤的保护作用.方法:将实验用昆明小鼠随机分为运动低剂量给药组、运动中剂量给药组、运动高剂量给药组、运动组和对照组,每组小鼠50只.运动低剂量给药组、运动中剂量给药组、运动高剂量给药组小鼠分别给予7.5、15.0、30.0 m g/k g剂量白藜芦醇灌服30 d,每天给药1次.对照组和运动组分别于相同时间给以生理盐水灌服30 d,给药1次/d.所有小组小鼠除对照组小鼠外每隔1 d给以力竭游泳训练1次,共30 d.实验结束后分别检测各组小鼠血清丙氨酸转氨酶(alanine aminotransferase,ALT)、天门冬氨酸转氨酶(aspartate aminotransferase,AST)及肝组织中一氧化氮(nitric oxide,NO)、一氧化氮合酶(nitric oxide synthase,NOS)、丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSHPX)、过氧化氢酶(catalase,CAT)、钠钾ATP酶(sodium potassium pump,Na+/K+-ATP)和钙镁ATP酶(calcium magnesium adenosine triphosphate synthase,Ca2+/Mg2+-ATP).同时,对小鼠肝脏组织进行HE染色.结果:相比较于对照组运动组的血清ALT、AST明显升高(P<0.05),肝组织MDA、NO、N O S明显升高(P<0.05),G S H-P X、N a+/K+-ATP和Ca2+/Mg2+-ATP显著降低(P<0.05),肝组织炎症及细胞凋亡情况明显比对照组严,重(P<0.05).相对于运动组小鼠,各给药组小鼠ALT、AST明显降低(P<0.05),肝组织MDA、NO、NOS明显降低(P<0.05),GSHPX、Na+/K+-ATP和Ca2+/Mg2+-ATP显著升高,(P<0.05),肝脏炎症情况情况轻于运动组,有浓度依赖关系.结论:白藜芦醇可以有效地保护力竭运动小鼠的肝功能. AIM:To investigate the protective effect of resveratrol on the liver of exhaustively exercised mice.METHODS:Kunming mice were randomly divided into a control group,an exercise control group,and low-,medium-,and highdosage resveratrol groups,with 50 mice in each group.All mice except those in the control group were given exhaustive swimming exercise every day for 30 d.The mice in the low-,medium-,and high-dosage resveratrol groups were given 7.5,15.0,and 30.0 mg/kg resveratrol by gastroenteric irrigation once a day for 30 days.The exercise control group and control group were given normal saline by gastroenteric irrigation once a day for 30 d.After the experiment,alanine aminotransferase(ALT)and aspartate aminotransferase(AST) in serum,and nitric oxide(NO),nitric oxide synthase(NOS),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(GSHPX),catalase(CAT),Na~+/K~+-ATP,and Ca^(2+)/Mg^(2+)-ATP in liver tissue were detected.Liver tissues were also stained with HE and subjected to microscopic evaluations.RESULTS:Compared with the control group,ALT and AST in serum,and MDA,NO,and NOS in hepatic tissue increased significantly(P 0.05),GSH-PX,Na~+/K~+-ATP and Ca^(2+)/Mg^(2+)-ATP in hepatic tissue decreased significantly(P 0.05),and hepatic inflammation and cell apoptosis were significantly more severe in the exercise control group.Compared with the exercise control group,ALT,AST,MDA,NO,and NOS decreased significantly(P 0.05),GSH-PX,Na~+/K~+-ATP and Ca^(2+)/Mg^(2+)-ATP significantly increased(P 0.05),and hepatic inflammation and cell apoptosis were significantly milder in the three resveratrol groups;these effects were concentration dependent.CONCLUSION:Resveratrol can effectively protect the liver of exhaustively exercised mice.
