摘要
目的:研究红景天胶囊的抗疲劳、抗缺氧作用,探索斑马鱼在中药药效筛选和中药毒性评价的应用可行性。方法:设置0.01、0.02、0.04、0.08、0.16mg·L-15个药物剂量组,将斑马鱼用药物处理5d,测定其游泳力竭时间、低氧耐受时间、低氧窒息后体内乳酸含量和乳酸脱氢酶(LDH)活性。结果:红景天在0.02mg·L-1时可明显延长斑马鱼低氧耐受时间(P〈0.05);0.01—0.16mg·L-1的红景天处理后5d后.斑马鱼的最大游泳速度均显著低于对照组(P〈0.05);药物浓度在0.04mg·L-1时,斑马鱼体内乳酸含量降低(P〈0.05);药物处理后,斑马鱼乳酸脱氢酶活性降低(P〈0.05)。结论:低浓度的红景天表现出抗缺氧的药性,可降低斑马鱼体内乳酸含量,表现出抗疲劳的效果;斑马鱼用于中药药物研究是可行的,需进一步探索药物处理时间和处理浓度,及其作用机理。
Objective:To investigate the effects of Rhodiola on the capacities of anti-fatigue and anti-hypoxia, to set up a method of judging the efficacy of traditional Chinese medicine using zebrafish, and to study the feasibility of using zebrafish model in Chinese medicine research. Methods:Zebrafish were randomly divided into five groups and administrated with different concentrations of Rhodiola solutions(0.01,0.02,0.04,0.08 and 0.16 mg . L-1) for 5 d. The maximal swimming speed, hypoxia tolerance time, body lactic acid content, and lactate dehydrogenase activity were measured. Results : The results showed that the Rhodiola of 0.02 mg . L -1 significantly extended the hypoxia tolerance time(P 〈0.05). 5 days treatment of Rhodiola with 0.01 -0. 16 mg. L-1 could reduce the maximum swimming speed of zebrafish ( P 〈 0.05 ). The Rhodiola of 0.04 mg. L- 1 reduced the lactic acid content ( P 〈 0. 05 ). The Rhodiola reduced the lactate dehydrogenase activity ( P 〈 0.05 ). Conclusion: The results showed that the low concentration of Rhodiola exhibits anti-hypoxia and anti-fatigue potency by reducing the lactic acid contents. Zebrafish can be used as experimental animal for Chinese drug research, but the treating time, drug dosage and its mechanism need to be further studied.
