摘要
目的:研究黄芪多糖(APS)对大鼠原代培养肝细胞抗氧化能力的影响作用。方法:采用"二步灌流法"制备大鼠原代肝细胞,给予40μmol/L的环磷酰胺造成脂质过氧化模型,然后给予三剂量(50、100及150μmol/L)的黄芪多糖,对照组给予相同体积的PBS,继续培养24 h,测定培养基上清生化、细胞内氧化及抗氧化指标,并评价黄芪多糖对环磷酰胺致毒性的保护作用。结果:相对于对照组,黄芪多糖各剂量组转氨酶(ALT,AST)及丙二醛(MDA)含量均明显下降(P<0.05),超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)活性均明显升高(P<0.05),且呈剂量依赖效应。结论:黄芪多糖对环磷酰胺引起的大鼠肝细胞脂质过氧化有一定的抑制作用。
Objective: To investigate the antioxidant effect of astragalus polysaccharides( APS) on primary cultured hepatocytes in rats. Methods: Rat primary cultured hepatocytes were prepared by two-step perfusion method. And the hepatocytes were given 40 μmol / L cyclophosphamide for establishing model of lipid peroxidation,then given three concentrations( 50,100 and 150 μmol / L) of APS. The model group was given the same volume of PBS,and cultured for continuous 24 h. The biochemical indicator of the supernatant and oxidant and antioxidant indicators of the cells in the medium were determined. And the protective effect of APS on cyclophosphamide-induced toxicity was evaluated. Results: Compared with the model group,the content of transaminase( ALT,AST) and malonaldehyde( MDA) significantly decreased( P〈0. 05),and the activities of superoxide dismutase( SOD) and glutathione peroxidase( GSH-Px) significantly increased( P〈0. 05),with a dose-dependent effect in each dose group of APS. Conclusion: APS had certain inhibitory effect on cyclophosphamide-induced lipid peroxidation in hepatocytes of rats.
