摘要
概述了低温对生物组织结构、生化反应及荧光特性的影响,总结了现有低温光学成像系统的结构和特点,从生物组织的三维结构信息和代谢、功能信息两个方面,对低温光学显微成像技术在生物学上的应用进行了回顾。认为随着技术的发展,低温光学显微成像技术将在获取大体积生物组织精细结构、探索与评价肿瘤治疗方法、研究代谢类疾病及神经系统疾病的代谢异常上发挥更大的作用。
Low-temperature optical microscopy is a novel biological research tool.It can keep in-situ structure and function states of biological tissue and organs by low temperature,and also provide three dimensional high-resolution structure and function in-formation in large range without optical limit.In this paper,the effects of low temperature on the structure and biochemical reac-tions of biological tissue and fluorescence are summarized,the configurations and features of current low-temperature optical ima-ging systems are reviewed,and the biological application of this technology are introduced from three-dimensional structure and metabolism as well as function information.
