胰岛素抵抗对卵巢癌细胞生物学功能及顺铂敏感性的影响

摘要 目的探讨胰岛素抵抗对卵巢癌细胞生物学功能及顺铂敏感性的影响。方法选取卵巢癌患者200例,根据是否患有2型糖尿病分为两组,胰岛素抵抗组83例、非胰岛素抵抗组127例。穿刺获取肿瘤组织和正常组织作为实验对象,采用免疫组化法测定IGF-1蛋白表达量,流式细胞术法测定IGF-1R蛋白表达量,MTT法测定不同浓度顺铂作用于肿瘤细胞的IC50(半数抑制浓度)值。结果 1胰岛素抵抗组IGF-1蛋白表达的HIS值为10.1±1.68,与非胰岛素抵抗组(6.5±2.14)比较,明显较高,差异有统计学意义(P<0.05);2胰岛素抵抗组IGF-1R蛋白表达量为(281.1±1.78),与非胰岛素抵抗组(264.6±5.32)比较,明显较高,差异有统计学意义(P<0.05);3胰岛素抵抗组不同浓度顺铂作用于肿瘤细胞的IC50值为(14.93±0.15),与非胰岛素抵抗组(2.41±0.08)比较,明显较高,差异有统计学意义(P<0.05)。结论胰岛素抵抗能够促进卵巢癌细胞增殖,降低卵巢癌细胞对顺铂的敏感性,对临床用药具有指导意义,值得临床推广。 Objective To study the effect of insulin resistance on the biological function of ovarian cancer cell and sensitivity to cisplatin.Methods 200 patients with ovarian cancer were selected,according to whether the patients had type 2 diabetes,were divided into 2 groups,the insulin resistant group 83 cases,and insulin non-resistant group 127 cases.Puncture for tumor and normal tissues was the experimental object,using immunohistochemical determination of IGF-1 protein expression,and IGF-1R protein expression was determined by flow cytometry method,MTT method for the determination of different concentrations of cisplatin on tumor cell IC50(half inhibitory concentration) values.Results 1The IGF-1 protein expression value of HIS of the insulin resistant group was 10.1 ± 1.68,which was significantly higher compared to insulin non-resistant group(6.5 ± 2.14),the difference was statistically significant(P〈0.05); 2The expression of IGF-1R protein in insulin resistant group was 281.1 ±1.78,which was significantly higher compared to insulin non-resistant group(264.6 ± 5.32),the difference was statistically significant(P〈0.05); 3The insulin resistant groups of different concentration of cisplatin to tumor cells IC50 values was 14.93 ±0.15,which were significantly higher compared to insulin non-resistant group(2.41 ± 0.08),the difference was statistically significant(P〈0.05).Conclusion Insulin resistance can promote the proliferation of ovarian cancer cells,decrease the sensitivity of ovarian cancer cells to cisplatin,has guiding significance for clinical medication,and it is worthy of clinical promotion.
