摘要
本试验采用响应面分析法,用Design Expert软件对马齿苋总黄酮的提取工艺参数进行优化,并对其大孔吸附树脂法纯化工艺进行了研究。结果表明:(1)最佳提取条件为:微波时间10 min、乙醇浓度75%、微波功率400W,在该最佳提取条件下总黄酮提取量为21.2592 mg/g,与理论预测值(21.8129 mg/g)接近;(2)确定HPD 600树脂的最佳纯化工艺条件:上样浓度为15 mg/m L,上样量为2 BV,洗脱剂浓度为80%乙醇,洗脱剂用量3 BV,在该条件下,纯化后的黄酮纯度为81.87%,约是纯化前(20.35%)的4倍。
In order to optimize the extraction and purification process of flavonoids from purslane,tile response sur- face methodology was used in the test, the optimum extraction process parameters were obtained by the software of Design Expert,and macroporous resin method was adopted on the purification process.The results was as follows: (1)the best ex- traction condition was the extracting time 10 min,the ethanol volume fraction 75%,and the microwave power 400 W.Under these conditions,the value of flavone extraction yield was 21.2592 mg/g approximating to the predicted value (21.8129 mg/g). Tile best purification condition of HPD600 macroporous adsorptive resins was sample concentration 15 mg/mL,volume 2 BV, tile elution concentration 80%,volume 3BV.The purified flavonoids was 81.87%,which was 4 times as much as before pu- rification of 20.35%.
