糖肾颗粒对糖尿病大鼠肾脏NF-κB和MCP-1表达的影响 被引量:10

Effects of Tangshen Particles on the expression of NF-κB and MCP-1 in diabetic nephropathy rats

摘要 目的:探讨糖肾颗粒对糖尿病大鼠肾组织核转录因子-κB(NF-κB)、单核细胞趋化因子1(MCP-1)表达的影响。方法:复制早期糖尿病肾病(DN)大鼠模型,将成模大鼠分为模型组,糖肾颗粒低、中、高剂量组和缬沙坦组,同时设正常对照组。观察大鼠一般状况、生化指标及肾组织病理,通过实时荧光定量PCR和Westernblot技术检测NF-κB和MCP-1 mRNA和蛋白表达,ELISA技术检测血中血栓素A2(TXA2)和前列环素(PGI2)含量。结果:模型组大鼠空腹血糖(FBG)升高,甘油三酯(TG)和胆固醇(TC)增加,尿微量白蛋白排泄率(U-alb)增加,内生肌酐清除率(Ccr)降低(P<0.01,P<0.05);TXA2、NF-κB、MCP-1明显升高,PGI2降低(P<0.05,P<0.01);经糖肾颗粒及缬沙坦干预后各项指标均有不同程度的好转。结论:糖肾颗粒通过降低NF-кB、MCP-1的表达抑制炎性反应从而对糖尿病肾脏损伤有保护作用。 Objective: To explore the effects of Tangshen Particles on the expression of NF-κB and MCP-1 in diabetic nephropathy(DN) rats. Methods: The early DN rat models were established, and then were randomly divided into DN model group; Tangshen Particles group with low, medium, high dosage, and Valsartan group. The normal group was set at the same time. The general condition, biochemical parameter and renal pathology were monitored. The mRNA and protein expression of NF-κB and MCP-1 were examined by real-time fluorescent quantitative(PCR) and Western blot method. The content of thromboxane A2(TXA2) and prostacyclin(PGI2) in serum were determined by ELISA methods. Results: In DN model group, fasting blood-glucose(FBG), triglyceride(TC), cholesterol(TG) and urinary albumin excretion rate were increased, and endogenous creatinine clearance rate(Ccr) was markedly decreased(P<0.01, P<0.05). TXA2, NF-κB and MCP-1 were significantly increased, and PGI2 was decreased(P<0.05, P<0.01). The above indicators were improved at Various levels by the intervention of Tangshen Particals and Valsartan. Conclusion: Tangshen Particles played a protective role on diabetic nephropathy by reducing the expression of NF- kappa B and MCP-1 to inhibit the inflammatory reaction.
