猪心脏脂肪酸结合蛋白基因多态性检测及与肉质性状的关联分析 被引量:8

摘要 随着人们生活水平的提高,对肉品质的要求也越来越高。在影响猪肉品质的众多因素中,遗传因素起到了很重要的作用。心脏脂肪酸结合蛋白基因(H-FABP)作为影响猪肉质性状的重要候选基因,曾在多个猪种上进行研究。本实验以3个安徽地方品种和1个杂交品种为材料(圩猪、皖南花猪、皖南黑猪、皖南黑猪与巴克夏的二元杂),将各品种的DNA分别混池,通过PCR扩增H-FABP基因的5′端调控区和全部4个外显子,进行测序检测SNPs,分别在5′端调控区和第2外显子各检测到1个SNP。这两个SNP位点与粗蛋白质、总脂肪、游离脂肪和16种氨基酸含量表型值关联分析表明,5′端调控区的SNP多态性与全部19个性状的关联结果未达显著水平(P>0.05),第2外显子处SNP多态仅与谷氨酸含量显著关联(P<0.05)。 With the improvement of people's living standards, pork quality is attracting more attention. Among the factors affecting pork quality, genetic factors play an important role. Heart fatty acid binding protein gene (H-FABP), as an important candidate gene for pork quality traits, has been investigated in several pig breeds. In this study, we used three Chinese indigenous pig breeds and one crossbreed originated from Anhui province (Wei pig, Wannan Hua pig, Wannan black pig and wannan black pig x Berkshire) to investigate the association of H-FABP with pork quality. DNA pool of each breed was constructed, and the 5~flanking regulatory region and all four exons of H-FABP were amplified by PCR. Two SNPs were finally detected, one is in its 51flanking regulatory region, and the other is in its exon The association studies showed that the SNP in exon 2 was significantly associated with glutamate trait only, while the SNP in the promoter region was not associated with protein content, total fat content, free fat content and 16 kinds of amino acid contents at all.
