摘要
目的探讨青蒿中有效成分的最佳萃取工艺条件及含量测定方法。方法运用Design-Expert软件对超临界二氧化碳(CO2)萃取方案进行优化筛选,采用高效液相色谱(HPLC)法测定青蒿萃取物中青蒿酸、东莨菪内酯、青蒿乙素和青蒿素等4种有效成分含量。结果最佳萃取工艺条件为:萃取温度50℃,萃取时间6 h,萃取压力4 5000 k Pa。青蒿酸、东莨菪内酯、青蒿乙素和青蒿素分别在0.045 6~0.273 6,0.004 9~0.029 2,0.066 6~0.392 0,0.201 0~1.206 0 mg·m L^-1浓度范围内线性关系良好。平均加样回收率分别为100.40%,100.81%,100.18%和101.60%。结论该工艺提取率高,HPLC能灵敏、准确地测定青蒿萃取物中4种有效成分的含量。
Objective To investigate the optimum extraction condition and content determination of effective components from Artemisia annua. Methods The Design-Expert software was applied to optimize supercritical CO2 extraction,and four components: artemisiaannua acid,scopoletin,arteannuin B and artemisinin in the extracts were determined by HPLC.Results The optimum extraction process was controlling extraction at 50 ℃,for 6 h,under pressure 45 000 k Pa. The linear ranges of artemisinic acid,scopoletin,arteannuin B and artemisinin was 0. 045 6- 0. 273 6,0. 004 9- 0.029 2,0. 066 6- 0. 392 0,0.201 0-1.206 0 mg·m L^-1,respectively.The average recoveries were 100.40%,100.81%,100.18%,101.60%,respectively.Conclusion This extraction process obtains high extraction rate. And the four effective components in Artemisia annua extracts can be determined sensitively and accurately by HPLC.
