摘要
目的:研究白藜芦醇对SOD1-G93A小鼠腰髓和运动皮层LC3B的影响。方法:36只SOD1-G93A小鼠随机分为3组,对照组、溶剂组、白藜芦醇组,每组12只,至症状早期(给药20~40天)或者终末期(给药60~80天)取材,利用激光共聚焦、免疫组织化学、Western-blot技术检测小鼠腰髓和运动皮层中LC3B的表达及LC3BⅠ/Ⅱ比值变化。结果:激光共聚焦技术及免疫组织化学技术检测发现症状早期白藜芦醇组(30mg/kg)和溶剂组小鼠腰髓运动神经元及运动皮层神经元与对照组相比LC3B表达增多;Western blot技术检测发现症状早期白藜芦醇组(30mg/kg/d)小鼠腰髓和运动皮层神经元中LC3BⅠ/Ⅱ比值分别为(1.20±0.29,1.27±0.30),与对照组相比(1.29±0.34,1.42±0.27)降低,但无统计学差异(P〈0.05)。终末期三组间小鼠腰髓和运动皮层LC3B表达及LC3BⅠ/Ⅱ比值无明显差异。结论:白藜芦醇对SOD1-G93A小鼠腰髓和运动皮层LC3B的影响微弱。
Objective: To study the influence of resveratrol on LC3B in lumbar spinal and motor cortex of SOD1 G93A mice. Methods: 36 SOD1 - G93A mice were randomly divided into control group, vehicle group and resveratrol group. Every group has 12 mice. Every mouse was treated differently until early-symptom or end-stage. According to the technology of electron microscopy confoeal microscopy and western blot , we can detect the different changes of LC3B expression and LC3B I / I/ ratio in different conditions of lumbar spinal and motor cortex. Results: Confocal laser technology and immunohistochemical technology detected, the LC3B expression increased in lumbar spinal motor neurons and motor cortex neurons of early symptoms resveratrol group (30 mg/kg/d) mice and vehicle group mice comparing with control group mice; Western blot technique detected, LC3B I / II ratio respectively isl. 20 ± 0.29 and 1.27 ± 0. 30 in lumbar spinal motor neurons and motor cortex neurons of early symptoms resveratrol group (30 mg/kg/d) mice, comparing with control goup mice 1.29 ±0.34 and 1.42 ±0.27. There is no significant difference ( P 〈 0. 05). The LC3B expression and LC3B I / Ⅱ ratio has no obvious difference between three groups of End-stage mice ( P 〉 0.05 ). Conclusion: The influence of resveratrol on LC3B is limited in the lumbar spinal and motor cortex of SOD1- G93A mice.
