裸燕麦β-葡聚糖调节血脂及改善肠源性内毒素血症作用的实验研究 被引量:5

The effects of naked oat β-glucan on the regulation of blood lipid and the improvement of intestinal endotoxemia

摘要 目的探究裸燕麦β-葡聚糖调节血脂过程中对肠道菌群的影响。方法用高脂饲料喂养金黄地鼠建立血脂异常模型,造模后将血脂异常鼠随机分为2组并分别以普通饲料、裸燕麦β-葡聚糖干预饲料饲养建立模型组(M)、干预组(OM),另设正常组(N)、预防组(OMN),以上各组动物每组12只,实验期90d。在干预初期、中期及末期检测各组血脂4项,同时进行粪便肠杆菌和双歧杆菌平板培养,末期测血清脂多糖(LPS)和炎症因子肿瘤坏死因子(TNF-α)、白介素-6(IL-6)。结果干预初期,OMN组的血清总胆固醇(TC)、总甘油三酯(TG)与低密度脂蛋白胆固醇(LDL-C)水平比N组低(P<0.05),OM组与M组血脂水平一致,干预末期,OM组血清TC、TG、LDL-C水平分别达到3.64mmol/L、2.52mmol/L、0.69mmol/L,相比造模时均显著下降(P<0.01),其中TC与LDL-C水平显著低于M组(P<0.01),此外,OM组肠杆菌与双歧杆菌比值在干预45d后即显著下降(P<0.01),并在90d时显著低于M组(P<0.01),同时,干预结束时OM组血清LPS水平为13.67μg/L,显著低于M组(P<0.01),OM组血清炎症因子IL-6为44.80ng/L、TNF-α为376.95ng/L,显著低于M组(P<0.01)。结论对血脂异常的个体,裸燕麦β-葡聚糖在有效调节血脂的同时可改善肠道肠杆菌与双歧杆菌比例并纠正菌群紊乱带来的肠源性内毒素血症,降低机体炎症水平。 Objective The aim of the study was to explore the effects of naked oatβ-glucan on gut microbiota(GM)while regulating blood lipid. Methods Dyslipidemia golden hamster models were established by feeding them with high-fat diet,then were randomly divided into model group fed with normal diet(group M)and experimental group fed with naked oatβ-glucan diet(group OM)for 90 days.Meanwhile,there were healthy hamsters fed with normal diet(group N)and naked oatβ-glucan diet(group OMN).Twelve hamsters were in each group.Serum lipid was measured and fecal Enterobacter and Bifidobacterium were cultured on the 1st,45 th and 90 th day,respectively.Also,serum lipopolysaccharides(LPS)and inflammatory factors were determined on the blood samples from the 90 th day. Results TC,TG and LDL-C levels of group OMN were significantly lower than group N at the beginning of the intervention(P〈0.05).After 90 days of intervention,they were 3.64 mmol/L,2.52mmol/L and 0.69 mmol/L in group OM,which significant decreased(P〈0.01).The levels of TC and LDL-C were lower than group M(P〈0.01).The ratio of Enterobacter to Bifidobacterium showed a significant decrease on the 45 th day(P〈0.01),and was much lower than that in group M by the end of intervention(P〈0.01).Meanwhile,the serum LPS,IL-6and TNF-αof group OM were all significantly lower(13.67μg/L,44.80ng/L,376.95ng/L)than that in group M at the end of intervention(P〈0.01). Conclusions Naked oatβ-glucan has a beneficial effect on blood lipid regulation and GM structure for individuals suffering from dyslipidemia,and on improving the intestinal endotoxemia and inflammatory conditions induced by dysbacteriosis.
