摘要
目的研究心脑通络液对动脉粥样硬化兔LDL-mRNA和VCAM-1mRNA表达的影响,探讨心脑通络液抗动脉粥样硬化作用。方法选取雄性新西兰兔40只,随机分为5组,即假手术组、模型组、心脑通络液液高剂量组(高剂量组)、心脑通络液低剂量组(低剂量组)、血脂康对照组,建立腹主动脉粥样硬化家兔模型。造模成功后,连续治疗2个月,RT-PCR法观察并检测不同时间点LDL-mRNA、VCAM-1mRNA的表达情况。结果高剂量组与血脂康组、低剂量组相比,LDL-mRNA表达升高,VCAM-1mRNA表达降低(P<0.05);高剂量组对动脉硬化家兔LDL-mRNA及VCAM-1mRNA表达的影响较血脂康组显著(P<0.05),且优于低剂量组(P<0.05)。结论心脑通络液可通过上调动脉粥样硬化兔腹主动脉细胞LDL-mRNA、下调腹主动脉细胞VCAM-1mRNA来改变动脉粥样硬化血管内皮基因表达失调,从而达到保护血管内皮细胞、抗动脉粥样硬化的作用。
Objective: To study the influence of Xinnaotongluo Liquid( XL) on LDL- mRNA and VCAM-1mRNA expression of atherosclerosis rabbits. Methods: 40 male New Zealand rabbits were randomly divided into five groups,namely,a sham operation group,a model group,a XL high- dose group,a XL low- dose group and a Xuezhikang control group to make abdominal aortic atherosclerosis rabbit models. After modeling,the rabbits were treated for continuous two months,and their LDL- mRNA and VCAM- 1mRNA expression at different time points were observed and detected by RT- PCR method. Results: Compared with that of the Xuezhikang group and the XL low-dose group,the LDL- mRNA expression of the XL high- dose group increased,but the VCAM- 1mRNA decreased( P〈 0. 05). The high- dose group had significant influence on the LDL- mRNA and VCAM- 1mRNA expression of the atherosclerosis rabbits( P 〈0. 05) and was better than the low- dose group( P 〈0. 05). Conclusion: Xinnaotongluo Liquid can mobilize aorta cell LDL- mRNA of atherosclerotic rabbits upwards and abdominal aortic cell VCAM- 1mRNA downwards to change the disorder of atherosclerotic vascular skin gene expression,thereby protecting vascular endothelial cells and resisting atherosclerosis.
