摘要
研究以一株从古井窖泥中分离筛选的枯草芽孢杆菌为出发菌株,进行发酵培养成分和发酵条件优化。与静置发酵相比,振荡培养可有效提高菌株发酵液中3-羟基-2-丁酮和2,3-丁二醇的产量;在振荡培养的基础上采用正交试验方法对发酵培养基和培养条件进行了优化。实验结果表明,枯草芽孢杆菌发酵培养的最佳条件为:3mmol/L Mg^(2+),15g/L蛋白胨,150r/min,32℃。与初始静置发酵条件相比,最优条件下3-羟基-2-丁酮、2,3-丁二醇(左旋)、2,3-丁二醇(内消旋)的产量分别提高了57.75%、1234.96%、163.33%。
This paper takes a Bacillus subtilis isolated and selected from Gujing pit mud as the original strain, and ts medium compositions and fermentation conditions were optimized. Compared with the rest of fermentation, shake-flask fermentation could effectively improve the flavor compounds in fermentation broth, and 3-hydroxy-2-butanone and 2,3-butanediol were the main compounds, and other by-products were less productive yield. To improve the 3-hydroxy-2-butanone and 2,3-butanediol-producing ability of Bacillus subtilis, orthogonal test was employed to optimize the medium compositions and fermentation conditions in shake-flasks fermentation experiments. The optimal culture medium and fermentation were follows: 3mmol/L Mg^(2+), 15g/L Peptone, 150r/min,32℃.Compared with the initial rest of fermentation,under the optimized condition the production of 3-hydroxy-2-butanone,(L)-2,3-butanediol and meso-2,3-butanediol increased by 57.75%, 1234.96% and 163.33%, respectively.
