摘要
目的通过激光诱导恒河猴眼底脉络膜新生血管(choroidal neovascularization,CNV)的形成,建立和人CNV类似的动物模型,用于定量评价Lucentis和Avastin治疗激光诱导引起的CNV的效果。方法采用眼科激光光凝仪围绕恒河猴黄斑中心凹进行光凝造模。光凝后21 d进行眼底荧光血管造影检查,根据荧光渗漏面积将动物分为空白对照组(生理盐水)、Avastin组(每眼1.25 mg)、Lucentis组(每眼0.5mg),均玻璃体内单次注射给药。给药后13 d、28 d进行眼底彩色照相、眼底荧光血管造影、光学相干断层扫描检查,给药后29 d摘取眼球进行病理学检查。结果与空白对照组相比,Avastin组在给药后13 d和28 d,荧光渗漏面积改善率分别为56.1%和82.0%,与空白对照组相比差异均有显著统计学意义(均为P<0.01),视网膜厚度改善率分别为14.2%和14.8%,与空白对照组相比差异均有显著统计学意义(均为P<0.01);Lucentis组在给药后13 d和28 d,荧光渗漏面积改善率分别为66.2%和75.6%,与空白对照组相比差异均有显著统计学意义(均为P<0.01),视网膜厚度改善率分别为19.8%和18.0%,与空白对照组相比差异均有显著统计学意义(均为P<0.01)。结论 Avastin和Lucentis对激光所致的猴眼底CNV均有明显的抑制作用。
Objective To establish an experimental model of choroidal neovascularization( CNV) through laser injury in the eyes of rhesus monkey,and quantitatively evaluate the therapeutic effects of Lucentis and Avastin on laser-induced CNV. Methods CNV model was induced by laser photocoagulation of the central macular fovea in rhesus monkeys,and confirmed by fluorescence fundus angiography( FFA) at 21 days post-exposure. According to the area of fluorescence leakage,monkeys were divided into three groups: Control group,Avastin( 1. 25 mg per eye) group and Lucentis( 0. 5mg per eye) group,and the drugs were single intravitreally injected. The fundus color photography,FFA and coherence tomography( OCT) were conducted at 13 days and28 days after giving drugs. Eyeballs were obtained and histopathology analysis was completed at 29 days after giving drugs. R esults Compared with the control group,the improvement rates of fluorescence leakage area at 13 days and 28 days after giving drugs in Avastin group were 56. 1 % and 82. 0 %,respectively,which in Lucentis group were 66. 2 % and 75. 6 %,respectively,there were significant differences compared with the control group( all P 〈0. 01). Compared with the control group,the improvement rates of retinal thickness at 13 days and 28 days after giving drugs in Avastin group were 14. 2 % and 14. 8 %,respectively,which in Lucentis group were 19. 8 %and 18. 0 %,respectively,there were significant differences compared with the control group( all P 〈0. 01). Conclusion Avastin and Lucentis can all obviously inhibit the laser-induced CNV in rhesus monkeys.
