摘要
目的:研究化浊解毒活血通络方对脑缺血再灌注损伤大鼠神经功能评分及Caspase-3表达的影响,探讨其神经保护作用机制。方法:采用改良线栓法建立大鼠脑缺血再灌注损伤模型。运用Longa法进行神经病学评分,用光镜观察脑组织形态学改变,用PCR法检测Caspase-3的表达。结果:模型组及各用药组均出现不同程度的神经功能缺损,模型组Caspase-3表达明显多于假手术组(P<0.05);与模型组相比,中药各组及阿司匹林组组织变性坏死程度轻、Caspase-3阳性细胞减少(P<0.05),中药高剂量组及阿司匹林组神经功能缺损评分显著降低(P<0.05);中药中、高剂量组和阿司匹林组效果优于中药低剂量组(P<0.05)。结论:化浊解毒活血通络方可以通过抑制Caspase-3的阳性表达来抑制细胞凋亡,从而发挥其神经保护作用。
Objective: To observe the influence of Huazhuo Jiedu Huoxue Tongluo Prescription(HJHTP) on scores of neuroiogic impairment and the expression of Caspase-3 in rats of cerebral ischemia reperfusion injury(CIRI), and explore its neuroprotective effect. Methods: Rat modes of CIRI were established through improved thread embolism method. The neuroiogic impairment was evaluated by Longa method. Pathological changes were observed with transmission electron microscope.The exepression of Caspase-3 protein was detected by PCR in the region of ischemia. Results: Neurologic impairment with different levels was observed in model group and medicine groups. The expression of Caspase-3 in the model group was increased than the sham operation group(P〈0.05). Compared with the model group, HJHTP groups and the aspirin group showed lighted tissue damage, and Caspase-3 positive cells were significantly less(P〈0.05). The scores of the high dose HJHTP group and the aspirin group were significantly lowered(P〈0.05). The effects of the high HJHTP dose group and middle dose HJHTP group were better than the low dose HJHTP group(P〈0.05). Conclusion: The neuroprotective effect of HJHTP on CIRI in rats may be involved in decreasing apoptotic neurons in cortex with inhibiting the the expression of Caspase-3 protein.
