摘要
目的研究+Gz暴露对冠脉不同程度狭窄模型猪血浆及血管巨噬细胞移动抑制因子(MIF)表达的影响。方法 25只巴马小型猪,在胸腔镜直视下手术丝线永久性结扎左前降支近端建立不同冠脉狭窄模型。各组模型猪进行+Gz暴露,并观察其最大+Gz耐受值。在最大+Gz暴露处理前后1 min留取静脉血并分离血浆,应用ELISA法检测MIF的含量。采用RT-PCR和免疫组化检测+Gz暴露后各组小型猪升主动脉MIF的表达情况。结果最大可耐受+Gz暴露后,各组模型猪MIF表达水平均较暴露前明显升高(P<0.05),且中、重度狭窄组模型猪血浆MIF浓度及升主动脉MIF表达水平均较假手术组明显升高(P<0.05),而轻度狭窄组则升高不明显(P>0.05)。结论 +Gz暴露对冠脉不同程度狭窄模型猪MIF的表达存在影响,且随狭窄程度加重,MIF表达增多。
Objective To investigate the effect of positive acceleration( + Gz) exposure on the expression of macrophage migration inhibitory factors( MIF) in swines with different degrees of coronary stenosis. Methods With 25 Bama miniature swines,the proximal left anterior descending branch( LAD) was ligated permanently using silk suture under direct vision of the thoracoscope to establish different models of coronary artery stenosis. + Gz exposure was performed in each group,and the maximum + Gz tolerance was observed. Blood was collected 1 minute before and after the exposure of the maximum + Gz acceleration tolerance in each group. Then,plasma was separated. The contents of MIF were measured by ELISA method. The expression levels of MIF were detected by RT-PCR and immunohistochemistry. Results After + Gz exposure,the expression of plasma MIF and MIF mRNA of the ascending aorta in the moderate and severe stenosis groups was significantly higher than that in sham operated group( P 〈0. 05),but not significantly elevated( P 〉0. 05) in the mild stenosis group. Conclusion + Gz exposure has effect on the expression of MIF in the models of coronary artery stenosis of different degrees,and the expression of MIF increases with the degree of stenosis.
