益母草碱对小鼠生理性月经样模型崩解期的作用 被引量:1

摘要 目的:研究益母草碱对小鼠生理性月经样模型的作用。方法:建立小鼠生理性月经样模型,将人工蜕膜化的假孕小鼠双侧卵巢切除,引发孕酮(P4)撤退,随后蜕膜化子宫内膜发生崩解坏死,模型小鼠出现月经样出血。在P4撤退前至撤退后24h,给予模型小鼠益母草碱,观察P4撤退后小鼠阴道出血状态,子宫大体形态以及组织形态学变化。结果:模型小鼠均在P4撤退后12~24h观察到阴道出血。与空白对照组相比,P4撤退后24h益母草碱药物处理组小鼠子宫颜色较深,子宫充血较显著。小鼠子宫腔上皮下的血管显著扩张,特别是腔上皮下血管扩张与红细胞渗出严重,且范围较对照组更广。通过统计学分析证实,益母草碱组充血面积高于对照组(P=0.011)。结论:提示益母草碱可导致小鼠子宫内血管扩张与红细胞渗出,其中的药理作用和分子机制有待进一步研究。 Objective:To study the effect of Leonurine hydrochloride(Yimucao)on physiological menstrual like model mouse.Methods:The menstrual like model mouse was established as follow,the artificially decidualized pseudopregnant female mice were received bilateral ovariectomy to trigger progesterone(P4)withdrawal,which resulted in endometrium breakdown,and the model mice appeared physiology menstruation.Leonurine was administrated to the model mouse from P4 withdrawal before to 24 hours after,so that the changes of mouse vaginal bleeding,general morphology and histomorphometry were observed.Results:Vaginal bleeding was observed in all mouse models from 12 to 24h after P4 withdrawal.At 24 hours,in Leonurine group,the uteri of mice were darker,and hyperemias of uteri were heavier than those in control group.Vasodilation in the endometrium breakdown area of the uterus was observed after Leonurine delivery,especially the area surrounding the luminal epithelium,which extended much wider compared to mice in control group.Statistical analysis further confirmed the relative hyperemia area was significantly higher in Leonurine group(P = 0.011).Conclusion:Leonurine hydrochloride is able to increase endometrial vasodilation and erythrocyte diapedesis in physiological menstrual like model mouse,however its pharmacological effect and functional mechanism need to be further explored.
