摘要
目的优选类球红细菌中辅酶Q_(10)的最佳超声提取工艺。方法采用HPLC法测定辅酶Q_(10)质量浓度,以菌体中辅酶Q_(10)的质量浓度作为响应值,在单因素试验基础上,选择超声波输出功率、超声波每次提取时间和超声波每次辐射时间这3个因素进行响应面试验设计。结果类球红细菌中辅酶Q_(10)的最佳超声提取工艺条件为:超声波输出功率为250 W,超声波每次提取时间为10 min,超声波每次辐射时间为8 s。在该条件下,辅酶Q_(10)质量浓度的预测值为11.535 1μg/m L,实测平均值为11.451 8μg/m L。结论本文所得工艺方法切实可行,为类球红细菌发酵生产辅酶Q_(10)的进一步研究提供参考。
Objective To study the extraction conditions of coenzyme Q10 from Rhodobacter sphaeroides by surface response methodology.Methods The concentration of coenzyme Q10 was set as response value,based on single factor experiments,and the response surface methodology was used to study the effects of ultrasonic power,ultrasonic extraction time at a time and ultrasonic radiation time at a time on the extraction rate of coenzyme Q10.The content of coenzyme Q10 was determined by HPLC.Results The optimal extraction conditions were as follows: ultrasonic power 250 W,ultrasonic extraction time at a time 10 min and ultrasonic radiation time at a time 8 s,under these conditions,the prediction value of coenzyme Q10was11.535 1 μg/m L and the measured value was 11.451 8 μg/m L.Conclusion The experimental process is feasible,which can provide technical reference for the further research of coenzyme Q10 in Rhodobacter sphaeroides.
