血管紧张素(1-7)对链脲佐菌素诱导的糖尿病大鼠胰岛β细胞凋亡的作用 被引量:1

摘要 目的探讨血管紧张素(1-7)[Ang-(1-7)]对链脲佐菌素(STZ)诱导的糖尿病大鼠糖代谢及胰岛β细胞凋亡的影响。方法健康雄性Wistar大鼠给予高脂高热量饲料喂养,8周后腹腔一次性注射STZ(30 mg/kg),建立2型糖尿病模型大鼠共20只,随机分为糖尿病对照组(D0组)和Ang-(1-7)干预组(D1组),同时设正常对照组(NC组)。D1组给予Ang-(1-7)300μg/(kg·d)皮下注射,余两组注射等体积生理盐水。干预共持续8周,监测空腹血糖(FPG),检测血清中空腹胰岛素(FINS)及AngⅡ的含量,计算稳态模型胰岛素抵抗指数(HOMA-IR),免疫组化法检测胰岛细胞内iNOS及Caspase 3的蛋白表达,实时荧光定量PCR法检测胰岛iNOS、bax、bcl-2及Caspase 3的mRNA表达。结果与NC组相比,D0组大鼠FPG、AngⅡ及HOMA IR明显升高,FINS浓度降低,胰腺iNOS、Caspase 3的蛋白表达分别增加了77.1%、1.03倍,iNOS、Bax、Caspase-3的基因表达水平分别增加3.4倍、2.9倍和4.6倍,Bcl-2基因表达降低了74.8%(P<0.05)。与D0组相比,D1组FPG、AngⅡ及HOMA-IR下降,FINS浓度升高,iNOS及Caspase-3蛋白表达分别降低了19.4%、37.2%,iNOS、Bax、Caspase 3的基因表达水平分别降低了67.6%、37.7%和39.7%,Bcl-2基因表达增加了81.2%(P<0.05)。结论 Ang-(1-7)可通过降低胰岛局部氧化应激,减少β细胞凋亡而改善STZ诱导的糖尿病大鼠胰岛分泌功能及胰岛素抵抗,发挥抗糖尿病作用。
