硫酸化相关酶在青少年特发性脊柱侧凸关节突软骨中的表达研究 被引量:1

摘要 [目的]分析硫酸化相关酶在AIS患者关节突软骨中的表达特点,为软骨细胞的力学研究和延缓脊柱侧凸进展提供参考。[方法]选取18例Lenke 1型AIS患者顶椎和下端椎凹、凸侧和对照组(15例)的下关节突软骨作为研究对象,固定脱钙后制成石蜡切片。免疫组化染色测定分析硫酸化反应相关酶PAPSS2、PAPST1和CHST1在Lenke 1型AIS患者顶椎和下端椎关节突软骨中的表达,利用Leica显微镜对免疫组化图像进行观察,用Image-pro Plus软件将每个视野中的着色面积、光密度以及目标数目转换成累积光密度值(IOD),代表相应酶的表达强度,最后进行统计学分析。[结果]PAPSS2主要分布在软骨细胞质和软骨囊基质;PAPST1特异性分布在软骨细胞质;CHST15主要分布在软骨细胞质和软骨囊基质。硫酸化反应相关酶PAPSS2、PAPST1和CHST15的表达转换成IOD有以下分布特点:对照组两侧的表达差异无统计学意义;顶椎凹侧高于凸侧,顶椎凸侧高于对照组,差异有统计学意义;顶椎高于同侧下端椎,差异有统计学意义;下端椎的凹凸侧的表达差异无统计学意义;下端椎的表达与对照组的表达比较差异无统计学意义。[结论]青少年特发性脊柱侧凸顶椎关节突硫酸化反应相关酶的表达增强与特发性脊柱侧凸进展相关。 [ Objective] To analyze the expression characteristics of sulfated -related enzymes in the region of apical and low end vertebral of adolescent idiopathic scoliosis, so as to provide theoretical basis for cytomechanics of the cartilage cell and the prevention or delay of the progress of scoliosis. [ Methods] The articular process cartukages of the concave and convex side of apical and low end vertebrae from 18 cases of Lenke type 1 adolescent idiopathic scoliosis and the control group ( n = 15 ca- ses) were collected respectively. The specimens were fixed, decalcified and embedded using standard procedures. The expression profiles of sulfation - related enzymes in articular process cartilages, both from the adolescent idiopathic scoliosis patients and the control group, were determined by IHC. The immunohistochemical images were observed by the Leica microscopy. The optical density in each colored area, and the number of targets were transformed into IOD, thereafter, to determine the expression inten- sity of corresponding enzyme with file help of Image - proPlus software. Then the data was statistically analyzed. [ Results ] The expression of the PAPSS2 was mainly found in the cytoplasm of cartilage cells and extracellular matrix of articular cartilages, the expression of the PAPSSTI was specially found in the cytoplasm of cartilage cells. The expression of the CHST15 was mainly found in the cytoplasm of cartilage cellsand extracellular matrix of articular cartilages. The expression of the sulfation - related en- zymes, PAPSS2, PAPST1 and CHST15 had no statistically significant difference between the bilateral sides of the eontroi group; the concave side of the apexes was higher than that of the convex side, the convex side was higher than that in the control group. The expression of the apex side was higher than that of ipsilateral low end vertebra. The comparsion of the convex side and the concave side of the low end vertebra had no statistically significant difference. The comparsion of the two sides of the low end verteura and the control group had no statistically signficant difference. [ Conclusion] The high expression of the sulfation -related enzymes PAPSS2, PAPSTI trod CHST15 in arlicular process cartilages of apical vertebral of adolescent idiopathic scoli- osis is corrclalcd with the progress of adolescent idiopathic scoliosis.
