摘要
目的探讨硫酸右旋糖苷(dextran sulfate,DS)对人胃癌细胞(BGC-823)低氧诱导因子-1α(hypoxia induced factor-1α,HIF-1α)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的抑制作用。方法将人胃癌BGC-823细胞分为对照组与实验组,在低氧条件下经不同时间培养后采用免疫细胞化学法及Western blot法检测HIF-1α、VEGF的表达水平。结果低氧条件下人胃癌BGC-823细胞中HIF-1α、VEGF的表达呈上升趋势,应用DS后实验组HIF-1α、VEGF的表达明显少于对照组,在12、24 h较为明显(P<0.001)。结论 DS可抑制人胃癌细胞中HIF-1α和VEGF的表达,并可能通过抑制其表达影响胃癌腹腔种植转移。
Purpose To investigate the inhibition of dextran sulfate (DS) on hypoxia induced factor-1 α( HIF-1α) and vascular endo- thelial growth factor (VEGF) in human gastric cancer BGC-823 cells. Methods In vitro, BGC-823 cells were divided into the control group and the experimental group under the condition of hypoxia. The expression levels of HIF-lα and VEGF was detected by immuno- cytochemistry method and Western blot at different times. Results The expression of HIF-lα and VEGF in human gastric cancer BGC-823 cells was increased under hypoxia condition and after treatment with DS, HIF-1α and VEGF protein expression was signifi- cantly less than that in control group, especially at 12 hours and 24 hours (P 〈 0. 001 ). Conclusion DS can inhibit the expression of VEGF and HIF-lα in human gastric cancer cells and have effect on peritoneal metastasis of human gastric cancer ceils.
