摘要
目的:探索蔓龙胆醇提物对小鼠急性酒精性肝损伤的保护作用及其安全性评价。方法:取昆明种小鼠60只,随机分为6组:正常对照组,模型对照组,蔓龙胆醇提物低、中、高剂量组(2.5、5、10 g/kg),联苯双酯阳性对照组0.2 g/kg;灌胃给药,每日1次,每周称一次体重,连续20 d,末次灌胃2 h后,除正常对照组灌等量蒸馏水外,其余各组灌服50%酒精12 ml/kg,禁食不禁水16 h后处死,按照试剂盒检测血清中谷天门冬氨酸氨基转移酶(AST)、甘油三酯(TG)、肝匀浆液总蛋白、谷胱甘肽(GSH)、丙二醛(MDA)。取昆明种小鼠40只,分为对照组和给药组,以最大给药浓度和最大给药容量灌胃给药,连续观察15 d后,测定其最大给药量倍数。结果:与模型对照组相比,蔓龙胆醇提物给药组可以降低血清AST、TG和肝匀浆MDA;给药组小鼠的最大给药倍数相当于成人临床用量428.8倍,安全性高。结论:蔓龙胆醇提物对酒精致肝损伤具有一定的保护作用,临床拟用药量安全。
Objective: Exploration of Gentianaceae alcohol extract on acute alcoholic liver injury in mice and the protective effect of safety evaluation. Methods: Take 60 mice are randomly divided into six groups: normal control group,model control group,Gentianaceae alcohol extract low,medium and high dose groups( 2. 5、5、10 g/kg),Bifendatatum positive control group 0. 2 g/kg; ig 1 time a day,according to weight,for 20 days,at the end of the time to fill the stomach after 2 h,in addition to the normal control group irrigation amount of distilled water,the rest of the group to fill take 50% alcohol 12 ml/kg,fasting and water for 16 h after death,according to the kits valley in serum aspartate amino transferase( AST),triglycerides( TG),liver and serum total protein and glutathione( GSH),malondialdehyde( MDA). Take 40 mice are randomly divided into two groups: control group and drug group,to the largest drug concentration and the maximum capacity to fill the stomach to medicine,continuous observation after 15 days,measure its maximum dosage ratio.Results: Compared with the model group,Gentianaceae alcohol extract dose group can decrease serum AST,TG and liver homogenate MDA; The maximum drug administration of the drug delivery group is equivalent to 428. 8 times of adult clinical dosage and high safety.Conclusion: Gentianaceae alcohol extract has a protective effect on liver injury induced by alcohol,the dosage of clinically safe.
