摘要
目的:研究不同白毛夏枯草提取物对肝癌细胞增殖的抑制作用及对其可能的作用机制进行初步的研究。方法:用白毛夏枯草水提取物(STW)、醇提物(CTW)、乙酸乙酯萃取部位(ZTW)分别作用于HepG2肝癌细胞,采用四甲基偶氮唑盐(MTT)法检测这3种提取物对HepG2肿瘤细胞增殖的抑制作用,观察不同浓度的白毛夏枯草水提物对肝癌细胞株HepG2的增殖影响。采用荧光定量PCR技术检测STW、CTW、ZTW处理后的肝癌细胞株中BAX和Bcl-2 mRNA的表达情况。结果:STW、CTW、ZTW均能显著抑制多种肿瘤细胞的增殖,与阴性对照组相比,其可在mRNA水平上抑制Bcl-2、同时促进BAX mRNA的表达,具有剂量依赖性。结论:白毛夏枯草提取物对肿瘤细胞具有明显的抑制作用,其抑瘤机制可能与抑制肿瘤细胞中Bcl-2、同时促进BAX mRNA的表达相关。
OBJECTIVE To investigate inhibitory effects of different extracts fromAjuga decumbens Thunb on cell proliferation and possible mechanisms.METHODS Methyl thiazolyl tetrazolium(MTT)assays were used to assess inhibitory effects of three extracts of Ajuga decumbens Thunb at different concentrations on HepG2 tumor cells.The three extracts were Ajuga decumbens Thunb straw water extract(STW),alcohol extract(CTW)and ethyl acetate extract(ZTW).BAX and Bcl-2 mRNA expression levels were analyzed by quantitative PCR.Three extracts of Ajuga decumbens Thunb significantly inhibited proliferation of HepG2 cells in a dose-dependent manner.RESULTS Compared with negative control group,these extracts could suppress expression of Bcl-2 on mRNA level,and increase BAX mRNA levels.CONCLUSION Ajuga decumbens Thunb extracts have obvious inhibitory effects on tumor cells,and the mechanism may be related to inhibition of Bcl-2 mRNA expression,and increased BAX mRNA expression in liver carcinoma cell line.
