西梅干预防大鼠草酸钙肾结石的实验研究 被引量:1

摘要 目的研究西梅干对乙二醇诱导的大鼠草酸钙肾结石形成的影响及其量效关系。方法 40只Wistar大鼠分为5组:A组空白对照组、B组为单纯乙二醇诱石组、C^E组为西梅干干预组[分别灌喂西梅干匀浆2、4、8mL/(kg·d)],每组8只。除A组外,余皆用含1%乙二醇去离子水于大鼠自由饮用。于实验前日和实验第4周末,分别检测大鼠尿钙、镁、草酸及枸橼酸浓度;血钙、镁、肌酐和尿素氮浓度;肾脏组织丙二醛(MDA)含量及总超氧化物歧化酶(T-SOD)活性;偏光显微镜观察HE染色肾脏组织草酸钙结晶形成情况;TUNEL法检测肾小管上皮细胞凋亡。结果三种剂量的西梅干均能明显减轻乙二醇所致的各项血尿生化指标和肾小管上皮细胞凋亡指数的改变,大幅增加尿镁浓度,明显减少乙二醇诱导的草酸钙肾结石的生成。结论三种剂量的西梅干均可明显干预乙二醇诱导的大鼠草酸钙肾结石的生成,且呈正相关的量效关系。 ObjectiveTo investigate the effects of prune intervention on ethylene glycol-induced calcium oxalate nephrolithiasis in rats and its dose-effect relationship. MethodsA total of 40 Wistar rats were divided into 5 groups:blank control group (group A), ethylene glycol-induced calcium oxalate group (group B), 2 mL/(kg·d) original homogenate of prune group (group C), 4 mL/(kg·d) original homogenate of prune group (group D), 8 mL/(kg·d) original homogenate of prune group (group D), with 8 rats in each group. Except for group A, the other groups were given 1% glycol deionized water to drink freely. Before and 4-week after the experiment, serum and urine concentrations of ionized calcium, magnesium, oxalic acid, and citric acid; blood concentrations of calcium, magnesium and creatinine were measured. After rats were sacrificed and renal tissues were obtained, the content of malondialdehyde (MDA) and activity of total superoxide dismutase (T-SOD) in renal tissues were determined. The formation of calcium oxalate crystals in renal tissues was observed with HE staining, and the apoptosis of renal tubular epithelial cells was detected by TUNEL method. ResultsAll of the three doses of prune could significantly reduce the change of biochemical index in blood and urine, and the renal tubular epithelial cell apoptosis index; increase the urinary magnesium concentrations; significantly reduce the calcium oxalate kidney stone formation in rats induced by ethylene glycol. ConclusionsAll of the three doses of prune can inhibit ethylene glycol-induced calcium oxalate nephrolithiasis in rats and there is a positive correlation between dose and effect.
