摘要
目的:探讨硫酸右旋糖苷(DS)通过调节HIF-1α(hypoxia induced factor,HIF-1α)的蛋白表达而抑制人胃癌细胞腹膜转移的相关机制。方法:(1)体外实验:培养BGC-823细胞,分别在培养液中加入DS和PBS,低氧培养不同时间,用免疫细胞化学和WB检测HIF-1α表达。(2)动物实验:构建胃癌腹膜转移模型,实验组与对照组分别注射DS和PBS。剖腹观察癌细胞在腹膜上种植转移情况;应用免疫组化和WB方法检测HIF-1α表达。结果:(1)体外实验:免疫细胞化学和WB结果显示实验组HIF-1α表达明显低于对照组。(2)动物实验:实验组瘤结节数明显少于对照组,免疫组化和WB结果显示实验组HIF-1α的蛋白表达明显低于对照组。结论:硫酸右旋糖苷可以调节人胃癌细胞HIF-1α的蛋白表达,其可能会通过下调HIF-1α的表达从而抑制人胃癌细胞大网膜种植转移。
Objective To explore the inhibition mechanisms of dextran sulfate (DS) on human gastric cancer cells peritoneal planting transfer by adjusting the protein expression of hypoxia induced factor-let. Method (1)Experiment in vitro: BGC-823 cells were cultured, then DS and PBS were added. Cells underwent hypoxic culturing of different periods. Immunoeytochemistry and WB were applied in testing Hif-let protein expression levels. (2)Animal experiment: an gastric peritoneal planting transfer model was established. The experimental and control groups received DS and PBS injections respectively. The distribution of tumour nodules in the celiac implantation was recorded. Immunohistochemistry and WB method were applied to detect Hif-let protein level. Results (1)Experiment in vitro: immunocytochemistry and WB results show that Hif-let protein level in the experimental group is significantly lower than in the control group. (2)Animal experiment: the number of planed tumour nodules of experimental group is obviously less than that of control group, immunohistochemistry and Western blot results show that the Hif-la protein expression of experimental group is significantly lower than the control group. Conclusion Dextran sulfate can regulate the Hif-let expression of human gastric cancer cells Hif-la through down-regulating expression of Hif-la, and thereby human gastric cancer cells peritoneal planting transfer was inhibited.
