黄芪皂苷对化疗贫血小鼠骨髓细胞c-Kit、SHP2、c-MPL等细胞因子的影响 被引量:8

摘要 目的:探讨黄芪皂苷对化疗贫血小鼠骨髓细胞INF-α、TGF-β、INF-γ和c-kit、SHP2、c-MPL细胞因子的影响。方法:从环磷酰胺所致的化疗贫血小鼠中,获取单个骨髓细胞,采用体外细胞培养方式培养小鼠骨髓细胞至第四代,并分成4组,分别为空白组、模型组、皂苷组和EPO组。采用ELISA法,观察黄芪皂苷对小鼠骨髓细胞中INF-α、TGF-β、INF-γ的影响;RT-PCR法观察黄芪皂苷对小鼠骨髓细胞中c-kit、SHP2、c-MPL的m RNA达的影响。结果:黄芪皂苷可以降低小鼠骨髓细胞中INF-α、INF-γ(P<0.05)的含量,提高TGF-β(P<0.05)的含量;还可以提高小鼠骨髓细胞中c-kit(P<0.05)的m RNA水平,降低SHP2(P<0.05)的m RNA水平,而对c-MPL(P>0.05)作用不明显。结论:黄芪皂苷能够改善TNF-α,TGF-β,INF-γ三种负性调节因子的水平,调节造血细胞的功能;上调c-kit,下调SHP2的基因水平,激活造血细胞的造血功能。 Objective:To explore the influence of Astragaloside on INF-α,TGF-β,INF-γ,c-Kit,SHP2 and c-MPL of the bone marrow cells in anemia model mice induced by chemotherapy.Methods:Chemotherapy anemia model was established by cyclophosphamide and the single bone marrow cell was obtained.The fourth-generation cells cultured in vitro were divided into four groups:normal group,model group,Astragaloside group and EPO group.The ELISA was used to test the concentrations of INF-α,TGF-β and INF-γ in mice bone marrow cells.The RT-PCR was applied to detect the m RNA contents of c-Kit,SHP2 and c-MPL of the bone marrow cells.Results:Astragaloside could decrease the concentrations of INF-α and INF-γ(P〈0.05),increase the contents of TGF-β(P〈0.05),raise the level of c-kit m RNA(P〈0.05) and reduce the assay of SHP2 m RNA(P〈0.05),but the variation of c-MPL contents was not obvious(P〉0.05).Conclusion:The levels of TNF-α,TGF-β and INF-γ were the three negative-regulate factors which were improved by Astragaloside and the? haematogenesis function was moderated.The hematopoietic system was activated by increasing contents of c-kit and lowering concentrations of SHP2.
