响应曲面法优化石榴皮总黄酮提取工艺 被引量:3

Optimized Extraction Process of Total Flavonoids from Pomegranate Peel by Response Surface Methodology

摘要 为获得超声波辅助提取石榴皮总黄酮的最佳工艺条件,在采用Plackett-Burman试验筛选出影响石榴皮总黄酮提取量的关键因素,通过最陡爬坡试验确定这些因素中心值的基础上,用Design-Expert 8.0.6软件中心复合试验(Central Composite Design,CCD)模块进行响应面分析。结果表明,初始温度和提取时间在研究所设水平范围内对石榴皮总黄酮提取影响不显著;以石榴皮总黄酮提取量最大值作为优化目标,在室温下提取10min,得到石榴皮总黄酮最佳提取工艺条件为:液料比(mL/g)82:1,乙醇分数66%,超声波功率154W;在此条件下进行5次平行验证试验,总黄酮平均提取量为165.10mg/g,达到理论预测值(171.22mg/g)的96.42%,表明该工艺条件稳定可靠,提取率高,可作为从石榴皮中大规模提取总黄酮的参考。 This paper investigated the optimum technological conditions for extracting total flavonoids from pomegranate peel by ultrasound-assisted extraction process.Key factors were selected by Plackett-Burman design,which could influence total flavonoids extraction efficiency from pomegranate peel.Steepest ascent experiment was conducted to determine the optimal level of these factors.The response surfaces were delineated based on the central composite design model in Design-Expert 8.0.6software.The results showed that the initial temperature and extraction time had no significant effect on the extraction of total flavonoids from pomegranate peel within the scope of the research institute.Extraction at room temperature for 10 min,the optimum extraction conditions were determined to maximize the extraction of total flavonoids from pomegranate peel:the liquid-to-solid ratio(mL/g),the proportion of ethanol and the ultrasonic power was 82:1,65%and 154 W,respectively.Under this condition,5parallel verification tests were carried out and the average value of total flavonoids extraction(165.09mg/g)reached 96.42%of the theoretical prediction value(171.22mg/g).It showed that the process was reliable with higher extraction rate,so it could be used as a reference to extract the total flavonoids from pomegranate pee on large scale.
