赤芍总苷对失血性休克大鼠血流动力学影响 被引量:11

Effect of total Saponins of Paeonia on hemodynamic indexes of rats with hemorrhagic shock

摘要 目的研究赤芍总苷(TSP)对失血性休克大鼠血流动力学的影响。方法用Wistar大鼠雌雄各半共24只,按照随机数字法分为4组:正常组、模型组、空白组和实验组,每组6只。4组大鼠麻醉后,都静脉注射肝素钠(2 mL·kg^(-1))全身抗凝,颈动脉插管放血,血压稳定30 min后,模型组、空白组和实验组放血至血压33.9 mm Hg,造成失血性休克模型。10 mL·kg^(-1)TSP用生理盐水配成0.5%的浓度后静脉给药,空白组给予等量的生理盐水。用生物信号采集分析系统观察大鼠的血压与心率,用自动血流变测试仪检测全血黏度、血浆黏度,红细胞变形指数、红细胞聚集指数、红细胞压积、全血还原黏度、全血相对黏度及血小板黏附率,观察TSP对失血性休克转归时血流动力学的调节作用。结果给与TSP后,实验组的全血黏度:200/s为(5.22±0.24)m Pa·s,30/s为(7.20±0.41)m Pa·s,5/s为(13.55±0.10)m Pa·s,1/s为(32.95±2.25)m Pa·s;血小板黏附率为(38.47±0.16)%,明显低于空白组的全血黏度:200/s(5.87±0.18)m Pa·s,30/s(8.46±0.23)m Pa·s,5/s(16.47±0.75)m Pa·s,1/s(41.77±1.61)m Pa·s和血小板黏附率为(39.08±0.24)%,差异有统计学意义(均P<0.05)。实验组的红细胞变形指数为0.79±0.01,明显高于空白组的0.71±0.02,差异有统计学意义(P<0.05),说明给药后红细胞变形能力明显增强。与休克时对比,用TSP复苏后,大鼠的收缩压为(106.00±13.24)mm Hg,舒张压为(76.99±13.41)mm Hg,平均血压为(80.33±8.82)mm Hg,差异有统计学意义(P<0.05)。结论 TSP能明显改善失血性休克时的血流动力学异常。 Objective To explore the effect of total of Saponins Paeonia ( TSP ) on hemodynamics in rats with hemorrhagic shock. Methods Male and female half, a total of 24 choose Wistar rats were randomly divided into normal group, model group, blank group and experimental group with 6 rats in each group. The rats were given intra- venous injection of hepariin sodium(2 mL· kg^-1) for body anticoagula- tion, followed by blood loss from carotid until model group, blank group and experimental group blood pressure at 33.9 mmHg and 30 minutes alter stability of blood pressure to make the models of hemorrhagic shock. The rats in the experimental group ( TSP 10 mL · kg^- 1 ) were intravenous injected with the solution diluted with saline. The rats in the blank group were given the same does of saline. At the same time, the blood pressure, heart rate were observed by biological signal acquisition and analysis system. The whole blood viscosity, plasma viscosity, erythrocyte deformation index, erythrocyte aggrega- tion index, red blood cell pressure, whole blood viscosity, whole blood relative viscosity, platelet adhesion rate were observed by automatic blood rheology tester. And the ratamics of hemorrhagic shock was observed. Results After treatment of TSP, the blood biscosity in experimental group at 200/s with (5.22 ± 0.24) mPa·s, at 30/s with (7.20 ± 0.41) mPa · s, at 5/s with (13.55 ± O. 10) mPa · s, at 1/s with (32.95 ± 2.25) mPa·s and platelet adhesion rate with (38.47 ± 0. 16) % were obviously lower than that of the blank group:blood biscosity at 200/s with (5.87 ±0. 18) mPa · s,at 30/s with (8.46±0.23) mPa · s,at 5/s with (16.47 ± 0.75) mPa · s, at 1/s with (41.77 ± 1.61) mPa · s and platelet adhesion rate with (39.08 ± 0.24)%, all about with statistical difference(P 〈 0.05 ). Meanwhile , the erythrocyte deformation in- dex in experimental group with 0.79± 0.01 were obviously higher than that of the blank group with 0.71± 0.02 and deformity ability of red blood cell was obviously increased ( P 〈 0.05). And compared with shock, after TSP recovery, systolic blood pressure with( 106.00 ± 13.24) mmHg, diastolic blood pressure with(76.99 ± 13.41 ) mmHg, mean pressure with (80.33 ± 8.82 ) mmHg in experimental group can get obvious improvement (P 〈 0.05). Conclusion TSP can obviously improve the abnonrmity of hemodynamics in rats with heamoregic shock.
