金线莲中槲皮素的超声提取及HPLC法测定 被引量:3

摘要 以金线莲(Anoectochilus roxburghii)为研究对象,采用超声波法提取,用高效液相色谱法测定了金线莲中槲皮素的含量。在单因素试验基础上,对乙醇体积分数、料液比、超声时间、超声功率4个因素进行了4因素3水平的正交试验。结果表明,槲皮素在0.000 5-0.010 0 mg/mL范围内呈良好线性关系,加样回收率为84.61%-85.90%。金线莲中槲皮素的最佳提取工艺条件为乙醇体积分数80%、料液比1∶25(g∶mL)、超声功率420 W、超声时间30 min。在此工艺条件下,金线莲中槲皮素的提取率为0.011 0%。试验对金线莲中有效成分含量的测定具有参考价值。 Quercetin in Anoectochilus roxburghii was ultrasonically extracted and detected by HPLC. On the basis of singlefactor experiment, an orthogonal test designed for the four factors including ethanol concentration, material-liquid ratio, ultrasonic time, ultrasonic power, and three levels was carried out. Results showed that the calibration curve of quercetin was well linear in the range of 0.000 5-0.010 0 mg/mL. The recovery was from 84.61% to 85.90%. The optimum extraction was carried out in 80%(V/V) ethanol for 30 min with the ultrasonic power of 420 W,and the material-liquid ratio of 1∶25(g∶mL).Under the technical conditions, the extraction yield of quercetin in Anoectochilus roxburghii was 0.011 0%. The experiment could provide valuable reference for the content determination of effective components in Anoectochilus roxburghii.
