退银汤对银屑病样小鼠模型NE、trappin-2、P-cad表达水平的影响 被引量:4

Effects of Tuiyin Decoction on expression levels of NE,Trappin-2 and P-cad in psoriasis like mouse model

摘要 目的:观察退银汤对咪喹莫特诱导的银屑病样小鼠模型中皮损组织和血清中性粒细胞蛋白酶(NE)、trappin-2、胎盘型钙黏蛋白(P-cad)等蛋白表达水平的干预作用,探讨退银汤治疗银屑病的作用机制。方法:使用5%咪喹莫特乳膏外涂小鼠背部脱毛区,制备银屑病样小鼠模型;72只小鼠随机分为正常对照组,模型对照组,退银汤低、中、高剂量组,消银颗粒组,每组12只,按照既定实验药量给各用药组灌胃给药,qd,连续14 d,正常对照组和模型对照组给予等容量的生理盐水;采用双抗体酶联免疫吸附法检测模型小鼠皮损组织及血清中NE、trappin-2、P-cad的含量。结果:模型组小鼠皮损组织及血清中NE、trappin-2、P-cad的含量均明显高于正常对照组(P<0.01);退银汤各剂量组能明显降低模型小鼠NE、trappin-2、P-cad的表达水平,与模型对照组比较均有显著性差异(P<0.05,P<0.01);退银汤中、大剂量组降低更为明显,与小剂量组比较均有显著性差异(P<0.05,P<0.01),并呈现良好的量效依赖关系;退银汤大剂量组皮损组织中的上述蛋白水平已接近正常对照组(P>0.05)。结论:退银汤能显著降低咪喹莫特诱导的银屑病样小鼠模型NE、trappin-2、P-cad等蛋白的表达水平,通过降低上述蛋白的表达水平来发挥调节免疫应答功能,可能是退银汤治疗银屑病的作用机理之一。 OBJECTIVE To observe the intervening effects of Tuiyin decoction on expression levels of NE,Trappin-2 and Pcad in skin lesions and serum of imiquimod induced psoriasis-like mouse models,and investigate the underlying mechanism.METHODS Psoriasis-like mouse models were established by applying 5% imiquimod cream to a hair-removal zone of mouse back.Seventy two mice were randomly divided into normal control group,model control group,low,middle and high dose groups of Tuiyin Decoction,and Xiaoyin granules group,with12 mice in each group.Mice in each group were treated by gavaging qd for 14 days consecutively.Mice in normal control group and model control group were treated with equal volume of saline.Concentration of NE,trappin-2,and P-cad in the skin lesions and serum of mouse model were detected by double antibody enzyme linked immunosorbent assay.RESULTS Concentrations of NE,Trappin-2 and P-cad in the skin lesions and serum of model group were all significantly higher than those in normal control group(P〈0.01).The expression levels of NE,Trappin-2 and P-cad of mice were significantly reduced in low,middle and high dose groups of Tuiyin Decoction compared with the model control group(P〈0.05 or P〈0.01).The reducing effects of middle and high doses of Tuiyin Decoction were more obvious compared with low dose group(P〈0.05 or P〈0.01),in a dose dependent manner.The protein expression levels of skin lesions in high dose group of Tuiyin decoction were close to those in normal control group(P〈0.05).CONCLUSION Tuiyin decoction can significantly reduce the expression levels of NE,Trappin-2 and P-cad in imiquimod-induced psoriasis-like mouse models,and regulate the immune functions by reducing expression levels of these proteins.It may be one of the mechanisms of Tuiyin decoction to treat psoriasis vulgaris.
