雷公藤多苷对变应性接触性皮炎小鼠干扰素诱导蛋白10及其受体表达的影响 被引量:4

摘要 目的观察雷公藤多苷对变应性接触性皮炎(ACD)小鼠体外培养脾淋巴细胞干扰素诱导蛋白10(IP-10)及其受体CXCR3表达的影响,进一步探讨雷公藤多苷治疗ACD的机制。方法以雷公藤多苷和RPMI-1640培养液,制备不同浓度雷公藤多苷培养液(高、中、低浓度组分别为5.0 mg/L、1.0 mg/L、0.5 mg/L),以不含有雷公藤多苷的培养液孔为空白对照。检测不同浓度雷公藤多苷溶液体外培养48 h后的ACD小鼠脾淋巴细胞中IP-10/CXCR3的表达,用逆转录聚合酶链反应(RT-PCR)检测IP-10,流式细胞术检测CXCR3。结果雷公藤多苷体外培养各组ACD小鼠脾淋巴细胞中IP-10 mRNA表达水平较之对照组受到抑制(P<0.05);与对照组比较,高、中浓度组表达水平降低明显(P<0.05),呈现出剂量依赖性。雷公藤多苷各浓度组体外培养的ACD小鼠脾淋巴细胞表面CXCR3分子表达与对照组比较均降低,但仅高浓度组抑制明显(P<0.05);高浓度组CXCR3分子表达水平低于低浓度组(P<0.05)结论雷公藤多苷可以抑制体外培养ACD小鼠脾淋巴细胞中IP-10 mRNA及其受体CXCR3的表达,对IP-10 mRNA的抑制呈现剂量依赖性,抑制或减轻炎症的产生,可能是雷公藤治疗ACD机制之一。 Objective In order to observe the effect of Tripterygium Glycosides on IP - 10/CXCR3 in ACD mouse, thus further to discuss the mechanism of this medicine in the treatment of contact dermatitis. Methods Different concentrations of Tripterygium wilfordii were prepared (the high, medium and low concentration groups were 5. 0 mg/L ,1.0 mg/L ,0.5 mg/L) , and the culture medium containing no tripterygium glyco-sides was used as blank control. The expression of IP - 10/CXCR3 in vitro culture of ACD mouse which incubation with Tripterygium Glycosides were detected by RT - PCR and flow cytometry. Results Compared with the control group, all of the three different concentration groups of Tripterygium Glycosides could remarkably suppress the expression of IP -10 mRNA in vitro culture Splenic lymphocyte of ACD mouse ( P 〈0. 05). Compared with the control group, the expression level of high concentration group was significantly reduced ( P 〈 0. 0 5 ) . Compared with the con-trol group, cultured ACD mouse spleen lymphocyte surface expression of CXCR3 of Tripterygium Glycosides groups decreased, but only the high concentration group decreased significantly ( P 〈0.05) . The CXCR3 expression level of high concentration group was lower than that of the lowconcentration group ( P 〈0? 05) ? Conclusion Tripterygium Glycosides can obviously suppress the expression of IP - 10/CXCR3 in vitro culture lymphocyte of ACD mouse, which maybe one of the mechanisms of Tripterygium Glycosides"treatment on ACD.
