抗人stathmin单克隆抗体和紫杉醇对肺癌细胞系QG-56的生长抑制和诱导凋亡作用

摘要 目的探讨抗人stathmin单克隆抗体(McAb)、紫杉醇(PTX)单用或联合应用对肺癌细胞系QG-56的生长抑制及诱导凋亡作用。方法以不同浓度的抗人stathmin McAb、PTX分别组成McAb组、PTX组和联合用药组,另设不加药的对照组,分别作用于QG-56细胞24、48、72、96h,观察细胞数量和形态的变化,MTT法检测各组QG-56细胞的增殖抑制情况,采用流式细胞仪检测各组QG-56细胞凋亡率。结果不同浓度的McAb组、PTX组和联合用药组细胞数量明显减少、形态不规则,部分细胞核固缩和胞质减少,而对照组细胞生长状态良好。抗人stathmin McAb、PTX单用和联合应用均能抑制QG-56细胞增殖,呈剂量-时间依赖效应,联合用药组细胞抑制率较McAb组、PTX组明显增大,差异有统计学意义(P<0.05),两药联用具有协同效应。抗人stathmin McAb、PTX单用与联用均能诱导QG-56细胞凋亡,联合用药组诱导凋亡作用更明显,差异有统计学意义(P<0.05)。结论抗人stathmin McAb、PTX单用与联用均能抑制QG-56细胞增殖,诱导其凋亡,两药联合作用于人肺癌QG-56细胞具有协同作用。 Objective To explore the effect of anti-human stathmin monoclonal antibody(McAb)and paclitaxel(PTX)in single or combined application on the growth inhibition and inducing apoptosis of lung cancer cell line QG-56.Methods The single drug group and the combined drug group were set according to the different concentrations of anti-human stathmin McAb and PTX respectively,while the control group received no drugs.The drugs were used on QG-56 cells for 24,48,72 and 96hours respectively to observe the changes of cell count and morphology.Methyl thiazolyl tetrazolium(MTT)method was used to detect the proliferation and inhibition of QG-56 cells,while flow cytometry(FCM)was adopted to detect the apoptotic rate of QG-56 cells.Results The cell count reduced significantly,the morphology was irregular,and the partial karyopyknosis and cytoplasm decreased in the single drug group and the combined drug group with different concentration of drugs,while cells had favorable growth conditions in the control group.Anti-human stathmin McAb and PTX in single or combined application could both inhibit the proliferation of QG-56 cells at a dosage-time-dependent manner.However,the combined drug group was significantly better in the inhibition rate of cells than the single drug groups(P 〈0.05),and the combination of two drugs had synergistic effect.Anti-human stathmin McAb and PTX in single or combined application could both induce the apoptosis of QG-56 cells,but the combined drug group was more significant in inducing apoptosis(P〈0.05).Conclusion Anti-human stathmin McAb and PTX in single or combined application can inhibit the proliferation and induce the apoptosis of QG-56 cells,but the combination of two drugs has a synergistic effect on human lung cancer QG-56 cells.
