摘要
【目的】建立沙葱组培快繁体系,为沙葱的规模化生产提供理论依据.【方法】以沙葱茎段为外植体,MS为基本培养基,探讨不同灭菌时间对种子污染率的影响,不同取材季节对沙葱愈伤组织诱导的影响,不同激素配比对愈伤组织、不定芽的诱导及生根的影响.【结果】种子的最佳灭菌时间为10min,污染率为6.7%.春季为沙葱的最佳取材季节,春季的沙葱茎段的出愈率、污染率分别为70.0%、15.7%.在2mol/L 6-BA和2mol/L NAA的激素配比下诱导的茎段出愈率最高,为72.5%,愈伤组织质地疏松;在1mol/L 6-BA和1mol/L NAA的激素配比下不定芽出芽时间12~15d,出芽率76%.在培养基1/2MS+0.5mol/L NAA上的生根率为88.9%、根长4.32cm、根粗0.86mm,为最适的生根培养基.【结论】沙葱种子的最佳灭菌时间为10min,茎段的最适的取材季节为春季,茎段脱分化最佳培养基为MS+2mol/L 6-BA+2mol/L NAA,不定芽最佳诱导培养基为MS+1mol/L 6-BA+1mol/L NAA,最适生根培养基为1/2MS+0.5mol/L NAA.
【Objective】To research the process of establishing rapid propagation system using stem segments of Allium mongolicumand provide theoretical basis for the large-scale production of Alliummongo licum.【Method】Using the stem of the Allium mongolicum as explants,and the MS as the minimal medium to discuss the influence of different sterilizing time on seed contamination rate,sampling seasons on callus induction and hormone combinations on callus induction,induction of adventitious buds and rooting.【Result】Optimum sterilizing time for seeds was 10 min and the contamination rate was 6.7%.Sampling in spring for Allium mongolicum stem section was the best,the callus rate and contamination rates was 70%and 15.7% respectively,which had extremely significant difference from those in summer and autumn.Stem callus rate was the highest under the hormone combination of 2mol/LNAA and 2mol/L6-BA reaching 72.5% and the callus texture was loose.Bud sprouting time of adventitious buds was for 12-15days under the combination of 1mol/LNAA and 1mol/L 6-BA,with budding rate of 76% differing significantly from that of other treatments.The rooting rate was 88.9%,root length was 4.32 cm and root thickness was 0.86 mm in medium of 1/2MS+0.5 mol/L NAA which was the best medium for rooting culture.【Conclusion】The results showed that the best sterilizing time for Allium mongolicumseed was 10 min,the best season for sampling stem segments was spring,the best callus medium for stem dedifferentiation of Allium mongolicum was MS+2mol/L 6-BA+2mol/L NAA.The optimal medium for adventitious bud induction was MS+1 mol/L6-BA+1 mol/L NAA.The most suitable rooting medium was 1/2MS+0.5mol/L NAA.
