毛木耳多糖的分离、分析及免疫药理活性的研究 被引量:43

摘要 毛木耳子实体经热水提取,Sevag法去蛋白,乙醇沉淀,得到毛木耳多糖(APP)。APP经Sephadex G-100柱层析,得到APP-A和APP-B两种多糖,经电泳鉴定均为单一区带,分子量分别为12万和7万,糖含量分别为68.33%和66.74%,糖醛酸含量分别为15.96%和15.58%。APP-A单糖组成摩尔比,岩藻糖:木糖:甘露糖:葡萄糖为0.05:0.18:3.20:1.00,APP-B为0.06:0.16:4.01:1.00。200~400 nm紫外扫描,无核酸和蛋白质吸收峰。4000~650 cm^(-1)红外扫描,呈多糖类特征吸收峰。原子吸收光谱分析表明,APP含多种对机体有益的微量元素。APP可增加小鼠脾指数,小鼠碳粒廓清率,增加巨噬细胞的吞噬功能,促进小鼠血清溶血素的形成,对大鼠红细胞有明显的凝集作用。 The polysaccharide was isolated from fruiting body of Auricularia polytricha Sacc (APP) with hot water and further purified with Sephedaex G-100. The two polysaccharides purified (APP-A and APP-B) were shown to be homogeneous by electrophoresis. The molecular weight of APP-A and APP-B was about 120 000 and 70 000 respectively, APP-A and APP-B contained sugar 68.33% and 66.74% reapcctively and uronic acid 15.96% and 15.58% respectively without protein and nucleic acid. Both APP-A and APP-B had polysaccharide characteristic absorbance by infrared scanning from 4000cm^(-1)~650cm^(-1). The molar ratios of sugars were also determined. APP-A, fucose : xylose : mennose : glucose=(0.05 : 0.18 : 3. 20 : 1.00); APP-B, fucose : xylose : mannose : glucose=(0.06 : 0.16 : 4.01 : 1.00). Atomic absorption spectra showed that APP-A and APP-B contained some trace elements which were healthful to organism. APP increased the spleen index, the charcoal clearance rate, the phagocytosis of peritoneal macrophages and production of hemolysis in mice. APP also promoted the agglutination of red cells in rat.
