摘要
目的初步探讨黄连素(Berberine)抑制胃癌细胞的分子机制。方法利用Real-time PCR检测黄连素对胃癌细胞BGC-823和SGC-7901增殖、凋亡、坏死、自噬及细胞连接相关基因表达水平的影响。结果黄连素处理胃癌细胞后,增殖相关基因IGFBP5、E2F6及MDM2表达水平降低,Notch2和JAG2表达水平升高,而PAK3表达水平无明显变化;凋亡相关基因Bcl-2表达水平降低,Caspase-1及Caspase-8表达水平升高;坏死相关基因RIPK1与RIPK3表达水平降低,自噬相关基因ATG7表达水平升高;细胞连接相关基因Ep CAM表达水平明显降低,E-Cadherin表达水平无明显变化。结论黄连素通过调控增殖、凋亡等相关基因的表达水平发挥抑制胃癌细胞的作用。
Objective To study the inhibition of berberine on gastric cancer cells. Methods The expression of proliferation, apoptosis, necrosis, autophagy and cell junction related genes in berberine-treated BGC-823 and SGC-7901 cells were detected by Real-time PCR. Results The expression of proliferation-related genes IGFBP5, E2F6 and MDM2 was decreased, the levels of Notch2 and JAG2 were highly increased, and the level of PAK3 did not change significantly in gastric cancer cells after treatment with berberine. Meantime, the expression of apoptosis-related gene Bcl-2 was decreased, but the levels of Caspase-1 and Caspase-8 were elevated. The expression of necrosis-related genes RIPK1 and RIPK3 was decreased, and the level of autophagy-related gene ATG7 was elevated. The expression of cell junction-related gene EpCAM but not E-Cadherin, was significantly decreased. Conclusion Berberine can inhibit gastric cancer cells by regulating the expression of proliferation and apoptosis genes.
