石榴皮、籽、花中总黄酮的含量比较及体外抗氧化能力研究 被引量:6

摘要 目的提取石榴皮、籽、花中的总黄酮并进行体外抗氧化能力的比较测定。方法以70%乙醇提取石榴皮、籽、花中总黄酮,采用直接显色法测定三者中总黄酮含量并测定石榴总黄酮对1,1-二苯基-2-三硝基苯肼(DPPH)自由基的清除能力、还原能力以及对羟自由基的抑制能力。结果1g石榴花中总黄酮含量为112.13mg,1g石榴皮中总黄酮含量为121.16mg,由于石榴籽与芦丁没有吻合的峰,故无法测量石榴籽中的黄酮含量。体外抗氧化实验中DPPH清除50%时所需要的石榴皮总黄酮的浓度为0.010 8mg/mL,石榴花总黄酮的浓度为0.015 1mg/mL。还原能力测定实验中,石榴皮总黄酮最大吸光度1.748,而同浓度维生素C仅为1.169;石榴花总黄酮最大吸光度1.732,维生素C为0.830;石榴皮总黄酮抑制能力对羟自由基抑制能力为355.7U/mL,同浓度维生素C溶液为273.2 U/mL,石榴花总黄酮为445.2 U/mL,同浓度维生素C溶液为239.1 U/mL。结论石榴皮中总黄酮含量>石榴花总黄酮含量。3个抗氧化试验结果显示石榴皮、石榴花总黄酮体外抗氧化能力为维生素C的2~3倍。 Objective To compare the flavone contents extracted from pomegranate peel,seed and flower respectively and observe the flavone′s antioxidative activity in vitro.Methods Extracted flavone from pomegranate peel,seed and flower respectively with 70% alcohol and measured flavone contents with direct colorimetry.Measured DPPH scavenging capacity and reducing capacity,and inhibitory ability of hydroxyl free radical.Results 121.16 mg flavone in 1 g pomegranate peel and 112.13 mg in 1 g pomegranate flower have been checked out.Peak of flavone in pomegranate seed was unable to be observed.In the experiment of DPPH scavenging,0.0108 mg/mL pomegranate peel flavone or 0.015 1 mg/mL pomegranate flower flavone were required to scavenge 50% DPPH.In the reducing capacity checking experiment,with equal concentration,the maximum absorbance of pomegranate peel was 1.748 abs,while that of Vitamine C was 1.169 abs;the maximum absorbance of pomegranate flower was 1.732 abs,while that of Vitamine C was 0.830 abs.The inhibitory activity of hydroxyl free radical of pomegranate flower flavone was 445.2 U/mL,while that of peel flavone was 355.7 U/mL.Conclusion The flavone in pomegranate peel is more than that in flower.Antioxidative activity of pomegranate′s flavone is as twice or three-fold as of VC in vitro.
