摘要
本实验旨在建立反相高效液相柱前衍生化色谱法测定壳聚糖喷雾剂盐酸氨基葡萄糖含量。实验采用Kromasil C18色谱柱,以乙腈-0.05%磷酸盐缓冲液为流动相,梯度洗脱,流速0.6 m L·min-1。实验结果表明,制剂中盐酸氨基葡萄糖衍生化后产生的2个异构体均达到良好分离,加样回收率平均值为90.09%,RSD为0.91%。本实验所建立的反相高效液相柱前衍生化色谱方法专属性强,可用于壳聚糖喷雾剂质量控制。
A pre-column derivation RP-HPLC method for determination of glucosamine hydrochloride content in chitosan spray was established. The target compound was analyzed by Kromasil C18 chromatographic column,acetonitrile-0. 05% phosphate buffer(16∶84) as mobile phase,gradient elution,0. 6 m L·min-1 as flow rate. The results showed that two isomers in glucosamine hydrochloride after derivatization were separated good. The average recovery was90. 09%,RSD was 0. 91%. In this experiment,the reversed-phase high-performance liquid pre-column derivatization chromatography method had strong specificity and can be used for the quality control of chitosan spray.
