摘要
目的:探讨温肾益气颗粒对慢性阻塞性肺疾病(COPD)大鼠气道PCNA的影响及相关机制。方法:40只SD雄性大鼠随机分为正常组、模型组、温肾益气颗粒组、雾化组。以气道滴注LPS联合烟熏方法制备COPD大鼠模型。观察肺组织病理改变,免疫组化检测气道PCNA表达,Western Blot观察肺组织中PCNA,p-AKT,TAKT,p-mTOR,T-mTOR的表达。结果:与空白组相比,模型组大鼠出现COPD病理变化,肺组织PCNA表达增强,p-AKT及p-mTOR表达增加(P〈0.05);与模型组相比,温肾组及雾化组大鼠COPD病理变化改善,PCNA表达降低,p-AKT及p-mTOR表达下降(P〈0.05)。结论:温肾益气颗粒降低COPD大鼠气道PCNA表达,其机制可能通过调节PI3K/AKT/mTOR信号通路实现。
Objective: To observe the effect and mechanism of Wenshen Yiqi Granule on the expression PCNA in rats with chronic obstructive pulmonary disease. Methods: Forty SD rats( male) were divided into norma group,model group,Wenshen Yiqi Granule treatment group and Atomization group. The rat model of COPD was established by intratracheal instillation of LPS and exposure to cigarette-smoking. Lung histopathological changes were observed. PCNA protein in lung tissue was measured by Immunohistochemical method. Western blotting was used to detect the expressions of PCNA,p-AKT,T-AKT,p-mTOR and T-mTOR. Results: Compared with the normal control group,there were remodeling of airway structure in three groups of COPD model of rats and the expressions of PCNA,p-AKT,p-mTOR in lung tissue of model group were higher significantly( P 〈 0. 05). Compared with the model group,Wenshen Yiqi Granule treatment group and Atomization group can alleviated the change of lung tissue and inhibit the expressions of PCNA,p-AKT and p-mTOR in lung tissue( P 〈 0. 05). Conclusion: Wenshen Yiqi Granule can inhibit the expression of PCNA in lung tissue. The mechanism may be realized by adjusting the PI3 K/AKT/mTOR signaling pathway.
