摘要
目的了解2013~2016年监测年度聊城市流感流行情况及甲型H3N2流感病毒HA1基因变异特征。方法采集哨点医院流感样病人的鼻咽拭子标本接种于MDCK细胞进行病毒分离及分型鉴定,采用一步法RT-PCR方法对随机抽取分离到的35株甲型H3N2流感病毒进行扩增,将扩增基因送生物公司进行HA1基因序列测定,利用DNAStar、MEGA软件进行同源性比对,并构建基因进化树。结果 2013年4月1日至2016年3月31日,共对2 137份标本进行MDCK细胞病毒分离,共检测出280株流感毒株,阳性率为13.10%。其中新甲型H1N1流感69株,占24.64%;甲型H3N2流感104株,占37.14%;B-Yamagata系流感86株,占30.71%;B-Vcitoria系流感26株,占9.29%。选取的30株甲型H3N2流感病毒进行测序,将其HA1基因分别与WHO推荐的当年度北半球流感疫苗株进行比对。结论 2013~2016流感监测年度聊城市甲型H3N2流感、B型流感和新型甲型H1N1流感病毒并存。
Objective To understand the epidemic situation of influenza virus and the genetic characterization of HA1 gene of influenza A viruses H3 N2 subtype in Liaocheng city,2013-2016. Methods The specimens of nasopharyngeal swabs were collected from influenza suspected cases of sentinel hospitals.Vaccination of the samples in MDCK cells to identify the virus separation.A one-step RT-PCR method was used to amplify 35 strains of influenza A viruses H3 N2 subtype.The amplification gene was sent to the biological company for HA1 gene sequencing.Using DNAStar and MEGA software for homologous comparison and genetic tree building. Results Vaccination of 2 137 specimens in MDCK cells to identify the virus separation from April 1,2013,to March 31,2016.A total of 280 strains of influenza were detected,with a positive rate of 13.10%.The influenza A viruses H1 N1 subtype was isolated in 69 strains,accounting for 24.64%.The influenza A viruses H3 N2 subtype was isolated in 104 strains,accounting for 37.14%.The influenza B viruses Yamagata subtype was isolated in 86 strains,accounting for 30.71%.The influenza B viruses Victoria subtype was isolated in 26 strains,accounting for 9.29%.The gene sequences were conducted on HA1 gene of 30 strains,and compared with vaccine strain recommended by WHO in the north globe. Conclusion The influenza A viruses H1 N1 subtype and the influenza A viruses H3 N2 subtype and the influenza B viruses Victoria subtype and the influenza B viruses Yamagata subtype exist in Liaocheng city,2013-2016.
