摘要
目的本研究拟运用RNA干扰肿瘤细胞CD133的表达,观察其能否提高结肠癌耐药细胞株对化疗药物的敏感性。方法构建CD133特异性RNA干扰慢病毒载体,转染结肠癌耐药细胞株,分析RNA干扰CD133的表达后对化疗耐药的逆转作用。结果成功构建了结肠癌耐药细胞模型及CD133特异性RNA干扰慢病毒载体。运用CD133特异性RNA干扰慢病毒载体转染人结肠癌耐药细胞株后,能够明显下调结肠癌耐药细胞株CD133的表达。细胞迁移实验表明,RNA干扰结肠癌耐药细胞株CD133的表达后,细胞迁移能力明显下降。RNA干扰结肠癌耐药细胞株CD133的表达后,其对5-FU的敏感性明显提高。结论运用RNA干扰下调CD133的表达后能够逆转结肠癌细胞对5-FU的耐药。
Objective To investigate the effects of down regulation CD133 expression by RNA interference on the chemosensitivity of chemotherapy resistance colon cancer cell line. Methods The recombinant lentivirus vectors targeting CD133 were constructed and transfected into chemotherapy resistance cells. The changes of CD133 expression by RNA interference ware analyzed,and the effect on the chemosensitivity ware further analyzed. Results The chemotherapy resistance colon cancer cell line and recombinant lentivirus vectors targeting CD133 were successful constructed. The recombinant lentivirus vectors targeting CD133 were transfected into the chemotherapy resistance colon cancer cells,and the down regulation of CD133 expression could be induced. The ability of cell invasion was measured using transwell chambers,and the invasion ability was obviously decreased after the RNA interference. After the down regulation of CD133 expression by the RNA interference,the chemosensitivity to 5-FU was significantly improved. Conclusion The down regulation of CD133 expression could be achieved by the RNA interference,and the drug resistance to 5-FU in chemotherapy resistance colon cancer cell line could be reversed.
