摘要
目的:采用溶剂诱导相变萃取-HPLC技术建立测定大鼠血浆中的沃诺拉赞浓度的方法。方法:色谱柱为Syncronis C_(18)色谱柱(250mm×4.6mm,5μm),柱温为40℃,流动相为20 mM NaH_2PO_4溶液(含0.1%甲酸)-乙腈(95:5,v/v),流速为1 mL·min^(-1),检测波长为230 nm。血浆样品采用溶剂诱导相变萃取法。结果:建立的方法专属性、线性、精密度与准确度、稳定性符合均生物样品测定要求。结论:溶剂诱导相变萃取-HPLC测定方法适用于大鼠血浆中沃诺拉赞浓度的测定。
Objective: To establish a solvent induced phase transition extraction - HPLC method for the determination of vonoprazan in rat plasma. Method: The chromatographic conditions were as follows: a Syncronis C18 chromatographic column (250mm×4.6mm, 5μm) was used at 40℃. The mobile phase consisted of 20 mM potassium dihydrogen phosphate buffer solution (containing 0.1% formic aeid)-acetonitrile (95:5, v/v ) with the flow rate at 1 mL· min^-1. The detection wavelength was 230 nm. The plasma samples were extracted by solvent induced phase transition extraction method. Result: The selectivity, linearity, precision, accuracy and stability of the established method were completely within the acceptance limits for biological samples. Conclusion: The solvent induced phase transition extraction - HPLC method was suitable for determination of vonoprazan in rat plasma.
