摘要
目的观察急腹症Ⅲ号对呼吸机相关性肺炎(ventilator-associated pneumonia,VAP)模型大鼠内毒素及细胞因子的影响。方法 80只雄性SD大鼠随机分为四组,急腹症Ⅲ号组:VAP模型大鼠每日灌胃急腹症Ⅲ号2 mL;谷氨酰胺组:VAP模型大鼠每日灌胃谷氨酰胺颗粒2 mL;急腹症Ⅲ号联合谷氨酰胺组:VAP模型大鼠每日灌胃急腹症Ⅲ号及谷氨酰胺颗粒各2 mL;安慰剂组:VAP模型大鼠每日灌胃生理盐水2 mL。给药5 d后采用酶联免疫吸附法检测大鼠血浆内毒素(ET)、白介素-2(IL-2)、白介素-6(IL-6)、核因子κB(NF-κB)水平。结果急腹症Ⅲ号组、谷氨酰胺组、急腹症Ⅲ号联合谷氨酰胺组大鼠血浆的IL-2、IL-6、ET水平均低于安慰剂组,谷氨酰胺组、急腹症Ⅲ号联合谷氨酰胺组大鼠血浆的NF-κB水平低于安慰剂组,急腹症Ⅲ号联合谷氨酰胺组IL-2、ET水平均低于急腹症Ⅲ号组,差异均有统计学意义(P<0.05)。结论急腹症Ⅲ号及谷氨酰胺均可抑制呼吸机相关性肺炎模型大鼠内毒素及相关细胞因子。
Objective To evaluate the effect of acute abdomen Ⅲ on endotoxin and cytokine in rats with ventilator-associated pneumonia(VAP).Methods A total of 80 male SD rats were divided into four groups,namely,acute abdomen Ⅲ group:VAP models,receiving 2 mL acute abdomen Ⅲ by gastric perfusion directly every day;glutamine group:VAP models,receiving 2 mL glutamine by gastric perfusion directly every day;acute abdomen Ⅲ combined with glutamine group:VAP models,receiving 2 mL acute abdomen Ⅲ and 2 mL glutamine by gastric perfusion directly every day;placebo group:VAP models,receiving 2 mL saline by gastric perfusion directly every day.The rats were killed on the sixth day and enzymelinked immunosorbent assay(ELISA) was used to detect the interleukin-2(IL-2),interleukin-6(IL-6),endotoxin(ET)and NF-κB level.Results The IL-2,IL-6 and ET level in the plasma of acute abdomen Ⅲ,glutamine group and acute abdomen Ⅲ combined with glutamine group was lower than those in the placebo group.The difference was statistically significant(P〈0.05).The NF-κB level in the plasma of glutamine group,acute abdomen Ⅲ combined with glutaminegroup was lower than that in the placebo group.The difference was statistically significant(P〈0.05).The IL-2 and ET in the plasma in the acute abdomen Ⅲ combined with glutamine group was lower than that of acute abdomen Ⅲ group.The difference was statistically significant(P〈0.05).Conclusions Both acute abdomen Ⅲ and glutamine can inhibit endotoxin and cytokines in rats with VAP.
